摘要
作者用多种高敏感免疫组化染色法和肝癌单克隆抗体HAb18,对肝癌组织切片染色,在光镜和电镜两个水平上观察HAb18相应抗原的分布与定位.并用HAb18配对AFP和铁蛋白单抗在同一切片上进行双重标记染色.结果提示,HAb18相应抗原选择性地表达于肝癌组织,其阳性率为75％(39/52),正常肝组织为阻性,其它恶性肿瘤仅有少数交叉.免疫电镜定位于肝癌细胞膜、内质网及一些管泡状结构的单位膜上.双重标记染色表明HAb18—Ag不同于AFP和铁蛋白.3种抗体复染均不能互相抑制.进一步提示HAb18—Ag为不同于已知抗原AFP和铁蛋白的另一肝癌标志物.其性质已由作者进行的免疫化学工作得到证实.
The distribution and localization of a new anti -hepatoma monoclonal antibody HAb18 corresponding antigen was studied. The results showed that this antigen is selectively expressed in hepatoma, with a positive rate of 75% (39/52). The expression of this antigen in normal liver tissue and in malignancies is scanty. The double staining of HAb18 in combination with either AFP or ferritin was performed. The results showed that these 3 antibodies did not inhibit each other on the same hepatoma sections because they had different staining patterns. Moreover, the HAb18 corresponding antigen was precisely localized subcellularly, by ultracryosectioning technique. A goat anti-mouse IgG colloidal gold (18 run)probe was used as an EM visualizer. The membrane,endoplasmic reticula, microvilli and some tubularvesicular structures were labelled with gold particles. The HAbl8-Ag was confirmed to be a membrane -localized antigen.
