摘要
立足天然药物化学实验教学体系与药学专业本科生实践需求,旨在优化桂圆壳多酚类物质超声提取工艺并检测其抗氧化活性.在超声功率500 W、温度80℃条件下,创新性引入响应面法优化工艺;并通过清除1,1-二苯基-2-三硝基苯肼(DPPH)、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)自由基清除实验进行活性检测.在t为50 min、乙醇体积分数为54%、液料比为26∶1 mL/g,桂圆壳多酚提取率为0.754%;经抗氧化能力测定所示,当桂圆壳多酚的质量浓度达到20 mg/mL时,其对DPPH、ABTS自由基清除率分别为93.34%和91.03%,与维生素C的清除效果无显著差异,说明桂圆壳多酚具有较强抗氧化活性.本文实现科研与教学深度融合,推动多学科知识交叉应用,有效提升了学生的综合实践能力,为天然药物活性成分开发及高校实验教学改革提供了切实可行的实践范例.
Based on the experimental teaching system of natural product chemistry and the practical needs of undergraduate students majoring in pharmacy,this study aimed to optimize the ultrasonic extraction process of polyphenols from Dimocarpus longan and detect their antioxidant activity.Under the conditions of ultrasonic power of 500 W and temperature of 80℃,the response surface methodology was innovatively introduced to optimize the process.The antioxidant capacity was detected through the experiments of scavenging 1,1-diphenyl-2-picrylhydrazyl(DPPH)and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt(ABTS)free radicals.When t was 50 min,the ethanol volume fraction was 54%,and the liquid-to-solid ratio was 26:1 mL/g,the extraction rate of polyphenols from D.longan was 0.754%.The antioxidant capacity test showed that when the mass concentration of polyphenols from D.longan reached 20 mg·mL-1,the scavenging rates of DPPH and ABTS free radicals were 93.34%and 91.03%,respectively,with no significant difference compared to the positive control vitamin C.This indicates that this polyphenol component has excellent antioxidant activity.This study achieved a deep integration of scientific research and teaching,promoted the cross-application of multi-disciplinary knowledge,effectively enhanced the comprehensive practical ability of students,and provided a practical example for the development of active components of natural drugs and the reform of experimental teaching in colleges and universities.
