摘要
文章开发了一种合成5-苄基丙二酸亚异丙酯的新方法。该方法以溴化苄和丙二酸二乙酯为起始原料,在无水碳酸钾存在下,以聚乙二醇(PEG-400)为相转移催化剂合成了苄基丙二酸二乙酯;再经碱处理和酸化反应制得苄基丙二酸中间体;最后,该中间体与丙酮在乙酸酐和浓硫酸的协同作用下发生缩合反应,顺利获得目标产物。通过红外光谱与核磁共振谱对产物的结构进行了确证。该合成路线具有反应条件温和、原料来源广泛、操作简便及产率较高等优点,为5-单取代丙二酸衍生物的合成提供了有益的参考。
A novel synthetic method for 5-benzyl isopropylidene malonate was developed.This approach employed benzyl bromide and diethyl malonate as starting materials.Using polyethylene glycol(PEG-400)as a phase-transfer catalyst in the presence of anhydrous potassium carbonate,diethyl benzylmalonate was synthesized.Subsequent alkaline treatment and acidification afforded the benzylmalonic acid intermediate.This intermediate then underwent a condensation reaction with acetone under the combined action of acetic anhydride and concentrated sulfuric acid,yielding the target product.The structure of the product was confirmed by infrared spectroscopy and nuclear magnetic resonance spectroscopy.This synthetic route offers advantages including mild reaction conditions,readily available raw materials,simple operation,and high yield,thereby providing valuable reference for the synthesis of 5-monosubstituted malonic acid derivatives.
