低共熔溶剂提取玉米须多糖工艺优化及抗氧化活性分析

Optimization of Extraction Process of Corn Silk Polysaccharide by Deep Eutectic Solvents and its Antioxidant Activity Analysis

摘要 研究旨在获取低共熔溶剂(deep eutectic solvents,DESs)法提取玉米须多糖最优工艺,并对玉米须多糖抗氧化活性进行分析.以DESs(尿素与氯化胆碱物质的量比2∶1)为提取试剂,以单因素试验为基础,考察玉米须多糖得率受液料比、DESs浓度、提取温度以及提取时间等条件变化的影响,通过响应面试验优化玉米须多糖提取工艺,并分析其清除DPPH自由基及ABTS自由基的性能.提取玉米须多糖最优参数为:液料比21∶1、DESs浓度42%、提取温度71℃、提取时间47 min.在此提取条件下,玉米须多糖得率为(5.38±0.02)%.所得玉米须多糖浓度为0.30 mg/mL时,其DPPH自由基及ABTS自由基清除率分别达到(88.69±0.17)%和(94.53±0.16)%,玉米须多糖对DPPH自由基及ABTS自由基清除的IC50值分别为0.121 mg/mL和0.111 mg/mL.优化后的玉米须多糖提取参数可靠,所得玉米须多糖抗氧化活性表现突出,可以作为天然抗氧化剂开发利用. This study aimed to optimize the extraction process of polysaccharides from corn silk using deep eutectic solvents(DESs)and to evaluate the antioxidant activity of the extracted polysaccharides.Using DESs(urea and choline chloride at a molar ratio of 2:1)as the extraction solvent,a single-factor experiment was conducted to investigate the effects of liquid-to-solid ratio,DESs concentration,extraction temperature,and extraction time on the yield of corn silk polysaccharides.Response surface methodology was used to optimize the extraction process,and the performance of the polysaccharides in scavenging DPPH and ABTS free radicals was analyzed.The optimal param⁃eters for extracting corn silk polysaccharides were:liquid-to-solid ratio 21:1,DESs concentration 42%,extraction temperature 71℃,and extraction time 47 minutes.Under these conditions,the yield of corn silk polysaccharides was(5.38±0.02)%.When the concentration of the obtained polysaccharides reached 0.30 mg/mL,the scavenging rates of DPPH and ABTS free radicals were(88.69±0.17)%and(94.53±0.16)%respectively.The IC50 values of corn silk polysaccharide for scavenging DPPH radical and ABTS radical were 0.121 mg/mL and 0.111 mg/mL,respectively.The optimized extraction parameters of corn silk polysaccharide are reliable,and the obtained corn silk polysaccharide exhibits outstanding antioxidant activity,which can be developed and utilized as a natural antioxidant.
