玉米须提取物对糖尿病足小鼠的干预作用 被引量:6

摘要 目的:通过研究糖尿病足小鼠模型给予玉米须提取物后血糖、胰岛素、体质量、内皮因子的变化水平,探索玉米须提取物对小鼠糖尿病足的干预效果。方法:150只小鼠适应性喂养1周后,随机选出30只小鼠作为空白对照组,其余进行糖尿病足造模。造模后随机分为模型对照组,玉米须提取液高、中、低剂量组,每组30只,空白对照组给予基础饲料喂养,其余4组给予高脂高糖饲料喂养至实验结束。第12周开始,进行糖尿病足模型造模,造模前禁食12 h以上,但可自由饮水,用浓度为0.1 mmol/L的柠檬酸(pH 4.2)缓冲液溶解链脲佐菌素(STZ),按照40 mg/(kg·d)的剂量腹腔注射,每日1次,连续5 d。测量小鼠6 h空腹血糖值,大于11.1 mmol/L显示造模成功。按照400 mg/kg、200 mg/kg、100 mg/kg的标准将玉米须提取物分别配成浓度为20 g/L、10 g/L、5 g/L的玉米须提取液,按照0.2 mL/10g给予高、中、低剂量组灌胃。持续给药30 d,给药第30天后1 h,禁食12 h以上,行小鼠尾采血进行指标检验。观察小鼠用药前后的血糖(FBG)、血清胰岛素(FINS)、体质量变化水平,内皮功能血清血栓素B_(2)(TXB_(2))、6-酮-前列腺素F-1α(6-K-PGF-1α)的指标含量变化。结果:与模型对照组比较,玉米须提取物高、中、低剂量组均具有降低FBG、TXB_(2)以及提高FINS、6-K-PGF-1α水平的作用(P<0.01),且呈现剂量依赖性,组间有差异(P<0.05),高剂量组要明显优于低剂量组,但是在改善体质量方面,玉米须提取液高剂量组改善小鼠的体质量有显著性差异(P<0.01),中、低剂量组有差异(P<0.05)。结论:玉米须提取物能够有效干预小鼠糖尿病足的血糖、胰岛素、体质量、内皮因子的变化水平。 Objective:To study the intervention effect of corn stigma extract on diabetic foot in mice by studying the changes of fasting blood glucose(FBG),fasting insulin(FINS),body weight and endothelial factors.Methods:After one week of adaptive feeding,30 mice were randomly selected from 150 mice as the blank control,the rest mice were established the model of diabetic foot and were randomly divided into the model group,the corn stigma extract groups of high-dose,medium-dose and low-dose,with 30 mice in each group.The blank control group was still fed with basic diet,and the other four groups were fed with high-fat and high-sugar diet until the end of the experiment.At the beginning of the twelfth week,the diabetic foot model was made.Before modeling,the mice were fasted for more than 12h,but they could drink water freely.The 0.1 mmol/L(pH4.2)buffer solution was used to dissolve Streptozotocin(STZ)for intraperitoneal injection at the dose of 40 mg/(kg·d),1 times a day for 5 consecutive days.The 6h fasting blood glucose of mice was measured,if the value was greater than 11.1 mmol/L,which indicated that the modeling was successful.The corn stigma extract groups of high-dose,medium-dose and low-dose were given corn stigma contract at the dose of 0.2 mL/10 g according to different concentrations of 20 g/L,10 g/L and 5 g/L respectively.The mice were intervened for 30 days,the mice were fasted for more than 12 hours after the last administration,and the sample blood was collected from mice tail for index detection.The changes of FBG,FINS,body weight,as well as serum thromboxane B_(2)(TXB_(2))and 6-keto-prostaglandin F-1α(6-K-PGF-1α)of endothelial function indexes were observed before and after the treatment.Results:Compared to those in the model group,FBG and TXB_(2) were decreased,FINS and 6-K-PGF-1αwere increased in the corn stigma contract groups of high-dose,medium-dose and low-dose after the treatment(P<0.01),there was a dose-dependent difference among the three groups(P<0.05),and the improvements in the high-dose group were significantly better than those in the low-dose group.Compared with the body weight in the model group,it was significantly improved in the high-dose group(P<0.01),the medium-dose group and the low-dose group after the treatment(P<0.05).Conclusion:Corn stigma extract can effectively interfere with the changes of FBG,FINS,body weight and endothelial factors in mice with diabetic foot.
