基于TCGA数据库分析长链非编码RNA SLC25A25-AS1与肺癌的相关性及其在肺癌诊断中的临床价值

Analysis of the correlation between long non-coding RNA SLC25A25-AS1 and lung cancer based on TCGA database and its clinical value in lung cancer diagnosis

摘要 目的基于癌症基因组图谱计划(TCGA)数据库分析长链非编码RNA(Lnc RNA)SLC25A25-AS1与肺癌的关系及其在肺癌诊断中的临床价值。方法从TCGA数据库下载肺癌及癌旁组织相关信息,分析肺癌中SLC25A25-AS1表达情况及其与肺癌临床因素的关系,评估SLC25A25-AS1在肺癌诊断中临床价值及其与肺癌细胞周期、凋亡调控基因表达相关性。结果SLC25A25-AS1在肺癌组织中的表达高于癌旁组织,差异有统计学意义(P<0.001)。不同SLC25A25-AS1表达水平的肺癌患者生存周期比较,差异无统计学意义(P>0.05)。不同大小、有无淋巴结转和远处转移的肺癌组织中SLC25A25-AS1表达比较,差异无统计学意义(P>0.05);不同Stage临床分期的肺癌组织中SLC25A25-AS1表达比较,差异无统计学意义(P>0.05)。SLC25A25-AS与肺癌肿瘤微环境中T细胞、滤泡辅助T细胞(TFH)、细胞毒性T淋巴细胞(CTL)、B细胞、巨噬细胞、自然杀伤细胞(NK)、中性粒细胞等多种免疫细胞及其亚群具有相关性(P<0.05)。SLC25A25-AS1+角蛋白19(KRT19)联合检测的诊断效能优于SLC25A25-AS1/KRT19单独检测。肺癌中SLC25A25-AS1与细胞凋亡调控基因BCL2(r>0.2,P<0.01)、侵袭调控基因MMP2/VIM等表达具有相关性(r<-0.2,P<0.01)。结论SLC25A25-AS1在肺癌的诊断中具有一定的临床价值,并且参与肺癌的肿瘤微环境免疫浸润,与肺癌中多种基因表达相关,有可能为肺癌诊疗策略中的辅助靶点。 Objective Based on the The Cancer Genome Atlas Program(TCGA)database,the relationship between long non-coding RNA(Lnc RNA)SLC25A25-AS1 and lung cancer and its clinical value in the diagnosis of lung cancer were analyzed.Methods Information related to lung cancer and adjacent tissues was downloaded from the TCGA database.The expression of SLC25A25-AS1 in lung cancer and its relationship with clinical factors of lung cancer were analyzed.The clinical value of SLC25A25-AS1 in the diagnosis of lung cancer and its correlation with the expression of lung cancer cell cycle and apoptosis regulatory genes were evaluated.Results The expression of SLC25A25-AS1 in lung cancer tissues was higher than that in adjacent tissues,and the difference was statistically significant(P<0.001).There was no statistically significant difference in the survival period of lung cancer patients with different expression levels of SLC25A25-AS1(P>0.05).There was no statistically significant difference in the expression of SLC25A25-AS1 in lung cancer tissues of different sizes,with or without lymph node metastasis and distant metastasis(P>0.05).There was no statistically significant difference in the expression of SLC25A25-AS1 in lung cancer tissues at different clinical stages(P>0.05).SLC25A25-AS was correlated with various immune cells and their subsets in the tumor microenvironment of lung cancer,including T cells,follicular helper T cell(TFH),cytotoxic T cell(CTL),B cells,macrophages,natural killer cells(NK),neutrophils,etc.(P<0.05).The diagnostic efficacy of the combined detection of SLC25A25-AS1 and cytokeratin-19 fragment(KRT19)was superior to that of the individual detection of SLC25A25-AS1/KRT19.The expression of SLC25A25-AS1 in lung cancer was correlated with that of apoptosis regulatory gene BCL2(r>0.2,P<0.01),invasion regulatory gene MMP2/VIM,etc.(r<-0.2,P<0.01).Conclusion SLC25A25-AS1 has certain clinical value in the diagnosis of lung cancer,participates in the immune infiltration of the tumor microenvironment of lung cancer,and is related to the expression of multiple genes in lung cancer.It may be an auxiliary target in the diagnosis and treatment strategies of lung cancer.
