CNN1作为膀胱癌早期诊断和预后潜在生物标志物的价值分析

摘要 目的旨在评估钙调蛋白1(calponin 1,CNN1)在膀胱癌患者中的表达特征及其预后价值,探讨CNN1与临床特征之间的相关性,并分析CNN1与免疫细胞的相互作用对肿瘤微环境的影响。方法通过癌症基因组图谱数据库获取433例膀胱癌样本的基因表达数据、临床病理信息及生存数据,分析CNN1与临床特征及患者预后之间的关系。根据CNN1表达水平进行分组,随后进行差异基因分析及富集分析;采用肿瘤免疫浸润分析预测免疫细胞的浸润水平差异,并分析其与CNN1的相关性。结果CNN1在多种肿瘤类型中表达异常,特别是在膀胱癌中显著降低,高表达CNN1的膀胱癌患者预后较差(P<0.05)。在临床特征分析中,CNN1与患者年龄和肿瘤分期呈正相关(P<0.05)。差异基因主要与细胞外基质和细胞因子受体相关。CNN1高表达引起初始B细胞和调节性T细胞的浸润增加,而与休眠自然杀伤细胞和滤泡辅助T细胞的浸润减少相关。结论CNN1在膀胱癌中总体表达较低,但与较高的临床分期相关。CNN1可作为膀胱癌早期诊断和预测预后的新型生物标志物。 Objective To evaluate the expression characteristics and prognostic value of calponin 1(CNN1)in patients with bladder cancer,while exploring the correlation between CNN1 and clinical features,to analyze the influence of the interaction between CNN1 and immune cells on tumor microenvironment.Methods Using The Cancer Genome Atlas database,gene expression data,clinical pathology information,and survival data for 433 bladder cancer samples were obtained.The relationship between CNN1 and clinical characteristics as well as patient prognosis were analyzed.Samples were grouped according to CNN1 expression levels,followed by differential gene analysis and enrichment analysis.The infiltration level differences of immune cells and analyzed their correlation with CNN1 were predicted by the tumor immune estimation resource.Results CNN1 exhibits abnormal expression across multiple tumor types,with particularly marked reduction observed in bladder cancer.Patients with high CNN1 expression demonstrate poorer prognosis(P<0.05).Clinical analysis revealed significant positive correlations between CNN1 expression and patient age as well as tumor stage(P<0.05).Differentially expressed genes were predominantly associated with extracellular matrix components and cytokine receptors.Elevated CNN1 levels correlate with increased infiltration of initial B cells and regulatory T cells,while being associated with reduced infiltration of dormant natural killer cells and follicle-associated helper T cells.Conclusion The overall expression of CNN1 in bladder cancer is low,and associated with a higher clinical stage.CNN1 can be used as a novel biomarker for the diagnosis and prognosis prediction of bladder cancer.
