Streptomyces sp. Y17A次级代谢产物的分离纯化及抑菌活性研究

摘要 本研究旨从渤海湾滨海盐沼湿地土壤分离的Streptomyces sp.Y17A菌株发酵液中分离得到具有良好抑菌活性的化合物。通过琼脂打孔扩散法和菌丝生长抑制法对其发酵液乙酸乙酯粗提物进行活性检测,结果显示该粗提物具有广谱的抑菌活性。在100μg·mL^(-1)的浓度下,对4株靶标细菌(柑橘黄单胞菌柑橘亚种、辣椒斑点病黄单胞菌、解淀粉欧文氏菌和胡萝卜软腐果胶杆菌)的抑菌圈直径为链霉素对照的58.3~65.7%;与溶剂对照相比,对5株靶标真菌(苹果黑腐皮壳、高粱生平脐蠕孢、立枯丝核菌、茄链格孢和葡萄座腔菌)菌丝生长的抑制率为63.0~100%。采用色谱技术从中分离纯化出4种化合物,通过核磁共振波谱与质谱分析确定其结构分别为asenjonamide C(1)、吲哚-3-甲醛(2)、吲哚-3-甲酸(3)和去甲哈尔满(4)。4个化合物均首次从Streptomyces sp.Y17A菌株发酵液中分离得到。微量稀释法检测结果显示这些化合物有不同程度的广谱抗菌活性,其中化合物1活性最显著,对柑橘黄单胞菌柑橘亚种等5种植物病原细菌的MIC为6.25~12.5μg·mL^(-1),对高粱生平脐蠕孢等7种植物病原真菌的MIC为12.5~50μg·mL^(-1)。可见,asenjonamide C具有开发为广谱抑菌先导化合物的潜力。 This study aimed to isolate compounds with potent antimicrobial activity from the fermentation broth of Streptomyces sp.Y17A,a strain obtained from coastal salt marsh soil of Bohai Bay.The antimicrobial activity of the ethyl acetate crude extract from the fermentation broth was evaluated using both the agar well diffusion assay and mycelial growth inhibition method.Results demonstrated that the crude extract exhibited broad-spectrum antimicrobial activity.At a concentration of 100μg·mL^(-1),the extract displayed inhibition zones measuring 58.3~65.7%of the streptomycin positive control against four target bacteria(Xanthomonas citri subsp.citri,Xanthomonas euvesicatoria,Erwinia amylovora,and Pectobacterium carotovorum subsp.carotovorum).Moreover,the extract also showed 63.0~100%inhibition of mycelial growth against five target fungi(Valsa mali var.mali,Bipolaris cookei,Rhizocto-nia solani,Alternaria solani and Botryosphaeria dothidea)compared to the solvent control.Chromatographic separation of the extract yielded four compounds,whose structures were determined to be asenjonamide C(1),indole-3-carboxaldehyde(2),indole-3-carboxylic acid(3),and norharman(4)by combined nuclear magnetic resonance(NMR)and mass spectrometry(MS)analyses.All four compounds were isolated for the first time from the fermentation broth of Streptomyces sp.Y17A.All compounds exhibited broadspectrum antimicrobial activity in broth microdilution assays.Compound 1 exhibited the most potent efficacy,with minimum inhibitory concentration(MIC)values of 6.25~12.5μg·mL^(-1)against five phytopathogenic bacteria,including Xanthomonas citri subsp.citri,and 12.5~50μg·mL^(-1)against seven phytopathogenic fungi,including Bipolaris cookei.These results indicated that asenjonamide C represented a promising broad-spectrum antimicrobial lead compound.
