金银花多糖增强伪狂犬疫苗免疫效果的研究 被引量:1

Study on enhancement of pseudorabies vaccine immune efficacy by Lonicera japonica polysaccharides

摘要 试验旨在探究金银花多糖(LJP)对伪狂犬病疫苗免疫小鼠的免疫调节作用。选取SPF级小鼠180只,随机分为6组,每组15个重复,每个重复2只。空白对照组(BC组)不免疫且不添加LJP,免疫对照组(VC组)免疫但不添加LJP,黄芪多糖组(APS组)免疫并灌服600 mg/kg黄芪多糖,LJP低剂量组(LJP-L组)免疫并灌服200 mg/kg LJP,LJP中剂量组(LJP-M组)免疫并灌服400 mg/kg LJP,LJP高剂量组(LJP-H组)免疫并灌服800 mg/kg LJP。预试期2 d,正式试验期32 d。结果显示,与VC组相比,LJP-M组脾脏和肝脏指数,以及LJP-H组肝脏指数均显著升高(P<0.05)。LJP-M组血清中免疫球蛋白G(IgG)、免疫球蛋白G1亚型(IgG1)、免疫球蛋白G2a亚型(IgG2a)抗体含量均极显著升高(P<0.01),免疫球蛋白G2b亚型(IgG2b)抗体含量显著升高(P<0.05);LJP-H组IgG和IgG2a抗体含量显著升高(P<0.05),IgG1和IgG2b抗体含量极显著升高(P<0.01);LJP-L组IgG1抗体含量极显著升高(P<0.01)。LJP-M组血清中干扰素-γ(IFN-γ)、白细胞介素-2(IL-2)和白细胞介素-4(IL-4)含量显著升高(P<0.05),白细胞介素-10(IL-10)含量极显著升高(P<0.01);LJP-L组IL-2和IL-4含量显著升高(P<0.05),IL-10含量极显著升高(P<0.01);LJP-H组IL-4含量极显著升高(P<0.01),IL-10含量显著升高(P<0.05)。研究表明,LJP可通过协同诱导1型辅助性T细胞/2型辅助性T细胞(Th1/Th2)混合型免疫反应,显著增强伪狂犬病疫苗的免疫效果,其中添加400 mg/kg LJP的效果较好。 This experiment aimed to investigate the immunomodulatory effects of Lonicera japonica polysaccharides(LJP)on mice immunized with pseudorabies vaccine.A total of 180 SPF-grade mice were randomly divided into six groups,with 15 replicates per group and two mice per replicate.The blank control group(group BC)was not immunized and did not receive LJP,the vaccine control group(group VC)was immunized but did not receive LJP,the Astragalus polysaccharide group(group APS)was immunized and administered 600 mg/kg Astragalus polysaccharide,the LJP low-dose group(group LJP-L)was immunized and administered 200 mg/kg LJP,the LJP medium-dose group(group LJP-M)was immunized and administered 400 mg/kg LJP,and the LJP high-dose group(group LJP-H)was immunized and administered 800 mg/kg LJP.The pre-test period was 2 days,and the formal test period was 32 days.The results showed that compared with group VC,the spleen and liver indexes in group LJP-M and the liver index in group LJP-H were significantly increased(P<0.05).In the group LJP-M,the serum levels of immunoglobulin G(IgG),immunoglobulin G1 subtype(IgG1),and immunoglobulin G2a subtype(IgG2a)antibody were extremely elevated(P<0.01),and the level of immunoglobulin G2b subtype(IgG2b)antibody was significantly increased(P<0.05).In the group LJP-H,IgG and IgG2a antibody levels were significantly increased(P<0.05),and IgG1 and IgG2b antibody levels were extremely increased(P<0.01).In the group LJP-L,the IgG1 antibody level was extremely increased(P<0.01).The levels of interferon-γ(IFN-γ),interleukin-2(IL-2),and interleukin-4(IL-4)in the serum of the group LJP-M were significantly increased(P<0.05),while the level of interleukin-10(IL-10)was extremely increased(P<0.01),the levels of IL-2 and IL-4 in the group LJP-L were significantly increased(P<0.05),and the level of IL-10 was extremely increased(P<0.01),the level of IL-4 in the group LJP-H was extremely increased(P<0.01),and the level of IL-10 was significantly increased(P<0.05).The study shows that LJP can significantly enhance the immune efficacy of the pseudorabies vaccine by synergistically inducing a mixed type 1 helper T cell/type 2 helper T cell(Th1/Th2)immune response,with the addition of 400 mg/kg LJP showing a better effect.
