金银花多糖对犬抗氧化、血液指标及疫苗免疫效果影响的研究

Effects of lonicera japonica polysaccharides on antioxidant capacity,blood indicators and vaccine immune efficacy in dogs

摘要 文章旨在给金银花多糖(LJP)在宠物食品中的应用提供借鉴,本研究选取50只6周龄的比格犬,随机分为5组,CK饲喂基础饲料,试验组LJP1、LJP2、LJP3和LJP4分别在基础饲料中添加0.1%、0.2%、0.4%和0.8%的LJP,预试期7 d,正试期42 d,试验结束后检测血清抗氧化水平及血液指标,分别于免疫前(0 d)及免疫后第14、28、42天检测犬瘟热抗体(CDV-Ab)和犬细小病毒抗体(CPV-Ab)水平。结果表明:LJP3和LJP4的活性氧(ROS)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和过氧化氢酶(CAT)活性显著高于其他组,丙二醛(MDA)含量显著低于其他组(P<0.05),且两组之间无显著差异(P>0.05)。与CK相比,LJP2的ROS、SOD、GSH-Px和CAT活性显著提高,MDA含量显著降低(P<0.05),LJP1各抗氧化指标均无显著变化(P>0.05)。添加LJP对红细胞(RBC)、血红蛋白(HGB)、红细胞压积(HCT)等指标均无显著影响(P>0.05)。同CK比,LJP3和LJP4的白细胞(WBC)和淋巴细胞百分比(LYM%)均显著提高(P<0.05),LJP2的WBC和LYM%有升高趋势,但未达显著水平(P>0.05)。免疫后第14天,LJP3和LJP4的抗体水平显著高于CK(P<0.05),且两组间无显著差异(P>0.05)。第28和42天,LJP2、LJP3和LJP4组抗体水平显著高于CK(P<0.05),其中LJP3和LJP4的抗体水平又显著高于LJP2(P<0.05),且两组间无显著差异(P>0.05)。综上所述,在犬粮中添加适量的LJP,能有效提高机体的抗氧化能力,提高白细胞和淋巴细胞数量,并显著提高犬瘟热抗体(CDV-Ab)和犬细小病毒抗体(CPV-Ab)水平,在本研究条件下,最适LJP添加剂量为0.4%。 This study aims to provide a reference for the application of honeysuckle polysaccharide(LJP)in pet food.Fifty 6-week-old Beagle dogs were selected and randomly divided into five groups.The CK group was fed a basal diet,while the experimental groups LJP1,LJP2,LJP3,and LJP4 were supplemented with 0.1%,0.2%,0.4%,and 0.8%LJP in the basal diet,respectively.The experiment included a 7-day adaptation period and a 42-day formal trial.After the trial,serum antioxidant levels and blood parameters were measured.Canine distemper antibody(CDV-Ab)and canine parvovirus antibody(CPV-Ab)levels were assessed before immunization(day 0)and on days 14,28,and 42 post-immunization.The results showed that the activities of reactive oxygen species(ROS),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),and catalase(CAT)in the LJP3 and LJP4 groups were significantly higher than those in other groups,while the malondialdehyde(MDA)content was significantly lower(P<0.05),with no significant difference between the two groups(P>0.05).Compared with CK,LJP2 showed significantly increased ROS,SOD,GSH-Px,and CAT activities and significantly reduced MDA content(P<0.05),while no significant changes were observed in any antioxidant indicators in LJP1(P>0.05).The addition of LJP had no significant effect on red blood cell(RBC),hemoglobin(HGB),or hematocrit(HCT)levels(P>0.05).Compared with CK,the white blood cell(WBC)count and lymphocyte percentage(LYM%)were significantly increased in LJP3 and LJP4(P<0.05),while LJP2 showed an increasing trend in WBC and LYM%,though not reaching statistical significance(P>0.05).On day 14 post-immunization,antibody levels in LJP3 and LJP4 were significantly higher than those in CK(P<0.05),with no significant difference between the two groups(P>0.05).On days 28 and 42,antibody levels in LJP2,LJP3,and LJP4 were significantly higher than those in CK(P<0.05),with LJP3 and LJP4 showing significantly higher levels than LJP2(P<0.05),and no significant difference between LJP3 and LJP4(P>0.05).In conclusion,adding an appropriate amount of LJP to dog food can effectively enhance antioxidant capacity,increase white blood cell and lymphocyte counts,and significantly improve canine distemper antibody(CDV-Ab)and canine parvovirus antibody(CPV-Ab)levels.Under the conditions of this study,the optimal LJP supplementation dose is 0.4%.
