摘要
为了解由青枯菌(Ralstonia solanacearum)引起的生姜青枯病生物防治策略,本研究从生姜根际土壤中分离对生姜青枯病具有生物防治潜力的拮抗菌株,探究其生物防治潜力及田间应用价值。从健康的生姜根际土壤中通过稀释、纯化分离得到菌株,采用平板对峙法筛选拮抗菌株,进行形态学观察、分子生物学鉴定。通过盆栽试验验证其对生姜青枯病的防治效果,同时检测菌株的生物防治相关性状。成功筛选出1株对青枯菌有明显抑制效果的菌株,通过形态学和多基因系统发育树分析鉴定为越南伯克霍尔德氏菌(Burkholderia vietnamiensis)B016,其生物防治性状表明菌株B016同时具有解磷、解钾和产嗜铁素能力。盆栽试验表明,B016对生姜青枯病有明显的防治效果,能降低青枯病发病率。该研究揭示越南伯克霍尔德氏菌B016能够有效抑制青枯病原菌,为开发生姜青枯病生物防治制剂提供优质菌种资源。
To develop biological control strategies against ginger bacterial wilt caused by Ralstonia solanacearum,this study isolated bacterial strains with biocontrol potential from the ginger rhizosphere soil and investigated their biocontrol efficacy and field application value.Bacterial strains were isolated from the rhizosphere soil of healthy ginger plants through dilution and purification.Antagonistic strains were screened using the dual-culture plate assay,followed by morphological characterization and molecular biological identification.Combined with pot experiments to verify their control efficacy against ginger bacterial wilt,the biocontrol-related traits of the strains were also assayed.A strain exhibiting significant inhibitory activity against R.solanacearum was successfully screened.Morphological observation and multi-gene phylogenetic analysis identified the strain as Burkholderia vietnamiensis B016.Assessment of its biocontrol traits demonstrated that strain B016 possesses phosphate solubilization,potassium solubilization,and siderophore production capabilities simultaneously.Pot experiments revealed that B016 significantly suppressed ginger bacterial wilt and reduced disease incidence.This study demonstrates that Burkholderia vietnamiensis B016 is an effective antagonist against the bacterial wilt pathogen,providing a promising microbial resource for the development of biocontrol agents against ginger bacterial wilt.
