油茶炭疽病病原鉴定研究

摘要 【目的】明确油茶炭疽病的主要病原菌,为科学防治该病害提供理论依据。【方法】对江西省南昌市江西农业大学油茶种植基地所采集的病叶样品进行病原菌的分离培养和鉴定,采用形态学和分子生物学的方法,结合病原菌的菌落形态、分生孢子形态、多基因系统发育树分析及致病性试验,明确其病原菌种类。【结果】基于ACT(肌动蛋白)、GAPDH(3-磷酸甘油醛脱氢酶)、ITS(内转录间隔区)的基因序列和形态学鉴定,确定油茶炭疽病的病原为果生炭疽菌和暹罗炭疽菌。【结论】明确了引起江西省南昌市江西农业大学油茶种植地油茶炭疽病的病原为果生炭疽菌和暹罗炭疽菌,为后续该病的分离鉴定与综合防控提供理论基础。 [Objective]This study aimed to identify the main pathogens of Camellia oleifera anthracnose,so as to provide a theoretical basis for the scientific prevention and control of this disease.[Method]Pathogens were isolated and identified from diseased leaf samples collected from the Ca.oleifera planting base of Jiangxi Agricultural University in Nanchang City,Jiangxi Province.Morphological and molecular biological methods were used,combining the colony morphology of pathogens,conidium morphology,multi-phylogenetic tree analysis,and pathogenicity tests to clarify the species of pathogens.[Result]Based on the gene sequence analysis of ACT(actin),GAPDH(glyceraldehyde-3-phosphate dehydrogenase),ITS(internal transcribed spacer)and morphological identification,the pathogens causing Ca.oleifera anthracnose were determined to be Colletotrichum fructicola and Colletotrichum siamense.[Conclusion]This study confirmed that the pathogens responsible for anthracnose in the Ca.oleifera planting area of Jiangxi Agricultural University in Nanchang City,Jiangxi Province,were C.fructicola and C.siamense.The results provide a theoretical foundation for the subsequent comprehensive prevention and control of this disease.
