摘要
【目的】为筛选高效的脂肪酶产生菌及其产酶工艺。【方法】采集被油污长期污染的土壤,经富集培养后,通过罗丹明B平板法进行菌株分离筛选;采用以花生油为唯一碳源的摇床发酵方法,研究所筛选出的菌株的产酶条件参数,并系统性优化各菌株产脂肪酶的条件。【结果】通过罗丹明B平板法筛选得到3株具有高产脂肪酶能力的菌株,并分别命名为LN2、Bk3与Bd4。经形态学及16S rRNA分子鉴定,LN2为铜绿假单胞菌(Pseudomonas aeruginosa),Bk3为越南伯克霍尔德氏菌(Burkholderia vietnamiensis),Bd4为土壤不动杆菌(Acinetobacter soil)。3菌株的最适产酶温度为30~40℃,最适发酵起始pH值为7.0。通过综合分析,培养48 h后,在发酵30℃、起始pH值7.0条件下LN2的酶活力为1.25 U/mL;在40℃、pH 7.0时Bk3的酶活力为0.85 U/mL;在30℃、pH值7.0条件下Bd4的酶活力为3.13 U/mL。【结论】3株脂肪酶产生菌在脂肪酶生产上均表现出优秀的潜能,尤其是Bd4,为脂肪酶工厂化应用提供候选菌种材料。
【Objective】The purpose of this experiment is to screen lipase producing bacteria and study their enzyme producing ability.【Method】The soil samples which had been contaminated by oil pollution for a long time,were enriched and cultured,and the strains were isolated and screened by Rhodamine B plate method.The shaking table culture with peanut oil as the only carbon source was used to explore the enzyme production parameters of the selected strains.The lipase production conditions of each strain were systematically optimized.【Result】Three strains with high enzyme production ability were obtained using Rhodamine B plate method and named as LN2,Bk3 and Bd4.Through by morphological and 16S rRNA molecular identification,LN2 was Pseudomonas aeruginosa,Bk3 was Burkholderia vietnamiensis and Bd4 was Acinetobacter soil.After 48 hours of cultivation,the optimal temperature for the three strains is 30~40℃,and the optimal culture pH is 7.0.Through comprehensive screening,the enzyme activity of LN2 was found to be 1.25 U/mL at 30℃and pH 7.0;the enzyme activity of Bk3 was 0.85 U/mL at 40℃and pH 7.0;and the enzyme activity of Bd4 was 3.13 U/mL at 30℃and pH 7.0.【Conclusion】The three lipase producing bacteria showed excellent potential in lipase production,especially Bd4,which provided candidate strain materials for the industrial application of lipase.
