摘要
黄瓜品种纯度鉴定是促进黄瓜生产持续稳定、高产的有效措施,对黄瓜种业发展和农业生产起着巨大的推动和保障作用。田间小区种植鉴定法易受环境影响,且周期长、效率低,无法达到快速鉴定的要求。SSR分子标记技术因其准确度高、实验周期短等特点,为品种纯度鉴定提供了一种更为便捷、高效的方法。以黄瓜新品种京研春秋绿2号及其亲本为材料,用92对SSR引物对其进行多态性筛选。结果表明,9对引物扩增得到的特异性片段呈现出丰富的多态性,亲本的带型特征在杂种后代上呈共显性分离,且具有互补性,构成了京研春秋绿2号的SSR指纹图谱,据此可以鉴定杂交种的纯度。其中8对引物扩增的条带清晰度高,易于分辨,利用这8对引物对京研春秋绿2号黄瓜种子进行检测,其鉴定结果与田间鉴定结果一致。
The purity identification of Cucumis sativus varieties is an effective measure to promote the continuous stability and high yield of Cucumis sativus production,and it plays a huge role in promoting and guaranteeing the development of Cucumis sativus seed industry and agricultural production.The field area planting identification method was more susceptible to environmental influences,and it had a long cycle and low efficiency,failing to meet the requirements of rapid identification.SSR molecular marker technology,with its high accuracy and short experimental cycle,provided a more convenient and efficient method for the purity identification of varieties.Polymorphism screening of the new Cucumis sativus variety Jingyan ChunqiulüNo.2 and its parents was conducted using 92 pairs of SSR primers.The results indicated that the specific fragments obtained from the amplification of the 9 pairs of primers presented rich polymorphism.The banding characteristics of the parents showed co-dominant separation in the hybrid offspring and were complementary,constituting the SSR fingerprint spectrum of Jingyan ChunqiulüNo.2.Based on this,the purity of the hybrid could be identified.Among them,the bands amplified by 8 pairs of primers were of high clarity and easy to distinguish.These 8 pairs of primers were used to test the seeds of Jingyan ChunqiulüNo.2 Cucumis sativus,and the identification results were consistent with the field identification results.
