一株二嗪磷降解菌Lactobacillus plantarum的分离、鉴定及其降解代谢途径

Isolation,identification and degradation metabolic pathway of a diazinon degrading bacterium Lactobacillus plantarum

摘要 [目的]分离筛选出一株二嗪磷降解菌株,完成菌株鉴定并对其降解特性和二嗪磷代谢途径进行分析。[方法]通过驯化和富集培养,从土壤样品中分离筛选二嗪磷降解菌株,基于形态学特征和16S rRNA基因序列分析,对菌株进行鉴定。采用单因素试验探究装液量、温度、pH及接种量对二嗪磷降解作用的影响。利用气相色谱-质谱(GC-MS)技术对二嗪磷的代谢产物进行鉴定。[结果]从土壤中分离得到一株可降解二嗪磷的细菌菌株TR6,经鉴定为植物乳杆菌Lactobacillus plantarum。进一步的降解条件优化结果显示,L.plantarum降解二嗪磷在250 mL三角瓶中最佳装液量为80 mL、最适接种量为5%、最适温度为37℃、最适初始pH为5.0。在降解二嗪磷过程中,共鉴定了2种代谢产物。[结论]L.plantarum可高效降解二嗪磷,经代谢后的产物毒性小于母体二嗪磷,且不产生高毒物质——二嗪酮,对环境有益,为二嗪磷农药的降解及降解机制提供了新的参考依据。 [Aims]This study aims to isolate,screen and identify a degrading bacterial strain,and analyze its degradation characteristics and metabolic pathways of diazinon.[Methods]Through acclimation and enrichment culture,a diazinon degrading strain was isolated and screened from soil samples.The strain was identified based on the morphological and 16S rRNA sequences analysis.The effects of liquid volume,temperature,pH and inoculation amount on the degradation of diazinon were investigated by single factor experiment.Gas chromatography-mass spectrometry(GC-MS)technology was used for identification of the metabolites of diazinon.[Results]A diazinon degrading bacterium TR6 isolated from soil was identified as Lactobacillus plantarum.Further optimization of degradation conditions showed that the optimal liquid volume for L.plantarum to degrade diazinon in a 250 mL Erlenmeyer flask was 80 mL,the optimal inoculation volume was 5%(by vol),the optimal temperature was 37℃,and the optimal initial pH was 5.0.During the degradation of diazinon,a total of two different diazinon metabolic products were identified.[Conclusions]L.plantarum can effectively degrade diazinon,and the toxicity of the products after metabolism is much less than diazinon,with no highly toxic substance-diazinone.These results provide new references for the degradation and degradation mechanism of diazinon.
