摘要
建立山药药材HPLC指纹图谱,为山药质量评价提供一种综合鉴定方法。采用HPLC法,色谱柱为中谱科技Polo-RP-C18(4.6 mm×250 mm,5μm);流动相为乙腈-0.05%磷酸溶液,梯度洗脱,流速0.5 mL/min;柱温25℃;检测波长210 nm。建立13批山药指纹图谱,采用相似度评价对数据进行分析。确定了7个共有峰,通过与对照品比对,指认了3个成分,分别为尿囊素、腺苷和苯丙氨酸,样品相似度均在0.95以上。该研究建立的指纹图谱方法简单可行,重复性好,可为山药药材的综合鉴定提供参考。
The study aimed to establish an HPLC fingerprint method for Rhizoma Dioscoreae and to provide a comprehensive approach for its quality evaluation.HPLC analysis was performed on a Polo-RP-C18 column(4.6 mm×250 mm,5μm),using acetonitrile-0.05%phosphoric acid solution as the mobile phase at a flow rate of 0.5 mL/min.The column temperature was maintained at 25℃,and the detection wavelength was set to 210 nm.Fingerprint chromatograms of 13 batches of Rhizoma Dioscoreae were generated and analyzed using the chromatographic fingerprint similarity evaluation system.A total of seven common peaks were observed across all batches,among which three peaks(allantoin,adenosine,and phenylalanine)were identified using reference standards.The similarity values of all samples were greater than 0.95.The established method is simple,feasible,and reproducible,offering a useful reference for the comprehensive quality evaluation of Rhizoma Dioscoreae.
