摘要 目的 建立HILIC-HPLC法测定山药中尿囊素的含量,用于中药材及中成药中尿囊素的含量测定。方法 色谱柱为Thermo Syncronis HILIC(250 mm×4.6 mm,5μm),流动相为乙腈-水(90:10),流速为1.0 mL min-1,柱温30℃,检测波长为191 nm。结果 尿囊素在16 min左右出峰。在0.100 4~1.004μg与峰面积线性关系良好(r=0.999 6),其平均回收率(n=6)为101.7%(RSD为1.9%)。结论 本法灵敏、准确、专属性强,能准确有效的控制山药中尿囊素的含量,并将出峰时间推迟至16 min,不易与其他峰重叠而满足中药材及中成药中尿囊素含量测定的需要。 Objective To establish an HILIC-HPLC method for allantoin in Dioscoreae Rhizoma. Methods The chromatographic procedure acetonitrile-water(90 :10) was carried out on a Thermo Syncronis HILIC(250 mm×4.6 mm,5 μm) column. The detection wavelength was 191 nm. The flow rate was 1.0 mL · min- 1 and the column temperature was 30 ℃. Results The peak time was 16 min. The linear range of allantoin was 0.100 4- 1.004 μg(r = 0.999 6), and the average recovery(n = 6) of allantoin was 101.7%(RSD was 1.9%). Conclusion The method is highly sensitive, reliable, accruate and can be applied to control the quality of Dioscoreae Rhizoma. The peak time has been postponed to 16 min, not easily overlaping with other peaks, and meeting the determination requirement of allantoin content in Chinese herbal medicine.
