基于网络药理学和分子对接探索黄芪桂枝五物汤治疗化疗诱导性周围神经病变的作用机制

Exploring the Mechanism of Huangqi Guizhi Wuwu Decoction Treating CIPN Through Network Pharmacology and Molecular Docking

摘要 目的:采用网络药理学方法系统分析黄芪桂枝五物汤治疗化疗诱导性周围神经病变(CIPN)的潜在机制。方法:从中药系统药理学数据库筛选复方活性成分及靶点,通过GeneCards数据库获取CIPN相关靶点,应用Cytoscape软件构建药物-成分-靶点互作网络,基于STRING数据库建立蛋白互作网络,对核心靶点进行GO功能和KEGG通路富集分析。通过AutoDock分子模拟软件进行分子对接验证成分与靶点的结合活性。结果:共筛选出40个活性成分及其对应的112个CIPN治疗靶点。功能富集分析表明黄芪桂枝五物汤通过调控IL-17通路、TNF信号转导、内分泌抵抗、MAPK级联反应、HIF-1信号系统、T细胞受体信号转导、Toll样受体通路以及Th17细胞分化等多种信号途径发挥对CIPN的治疗效应。分子对接结果表明代表性成分与关键靶点具有稳定结合活性。结论:黄芪桂枝五物汤通过多成分、多靶点、多通路协同作用治疗CIPN的机制,有助于深入理解黄芪桂枝五物汤治疗CIPN的潜在分子作用机制,为后续机制研究和临床应用提供理论基础。 Objective:To explore the mechanism of Huangqi Guizhi Wuwu Decoction treating chemotherapy-induced peripheral neuropathy(CIPN)using network pharmacology approaches.Methods:The bioactive constituents and their associated molecular targets of Huangqi Guizhi Wuwu Decoction were obtained from the TCMSP database,while disease-related targets for CIPN were acquired from GeneCards.Subsequently,an integrative network depicting herb-compound-target interactions was visualized employing Cytoscape software open-source bioinformatics software.A protein-protein interaction(PPI)network was established based on the STRING database,followed by analysis of protein interaction relationships.Gene Ontology(GO)annotation and KEGG pathway enrichment analyses were carried out on the common targets.The molecular interactions between active compounds and core targets were verified through AutoDock-based docking simulations.Results:A total of 40 active components and 112 potential therapeutic targets for CIPN were identified.Functional enrichment analysis showed that Huangqi Guizhi Wuwu Decoction exerts therapeutic effects on CIPN by regulating multiple signaling pathways,including IL-17 pathway,TNF signaling pathway,endocrine resistance,MAPK cascade reaction,HIF-1 signaling system,T cell receptor signaling pathway,Toll like receptor pathway,and Th17 cell differentiation.Molecular docking demonstrated stable binding interactions between representative components and core targets.Conclusion:This study elucidates the multi-component,multi-target,and multi-pathway synergistic mechanisms of Huangqi Guizhi Wuwu Decoction in treating CIPN,providing insights into its potential molecular mechanisms and laying a theoretical foundation for further experimental research.
