摘要
目的 建立UPLC-MS/MS法同时测定补阳还五汤中腺嘌呤、芍药苷、咖啡酸、绿原酸、苦杏仁苷、香兰素、毛蕊异黄酮葡萄糖苷、毛蕊异黄酮、槲皮素、山柰酚、芒柄花黄素、β-谷甾醇、豆甾醇、黄芪甲苷、阿魏酸、丹皮酚、羟基红花黄色素A的含量。方法 分析采用Kinetex XB-C_(18)色谱柱(100 mm×3 mm, 2.6μm);流动相水(含0.1%甲酸)-甲醇,梯度洗脱;体积流量0.4 mL/min;柱温40℃,电喷雾离子源;正负离子扫描;多反应监测模式。结果 17种成分在各自范围内线性关系良好(r>0.990 0),平均加样回收率87.2%~104.9%,RSD 0.76%~6.55%。结论 该方法简便快速,灵敏度高,专属性强,可用于补阳还五汤的质量控制。
AIM To establish a UPLC-MS/MS method for the simultaneous content determination of adenine,paeoniflorin,caffeic acid,chlorogenic acid,amygdalin,vanillin,calycosin-7-glucoside,calycosin,quercetin,kaempferol,formononetin,β-sitosterol,stigmasterol,astragalosideⅣ,ferulic acid,paeonol and hydroxysafflor yellow A in Buyang Huanwu Decoction.METHODS The analysis was performed on a 40℃thermostatic Kinetex XB-C_(18) column(100 mm×3 mm,2.6μm),with the mobile phase comprising of water(containing 0.1%formic acid)-methanol flowing at 0.4 mL/min in a gradient elution manner,and electron spray ionization source was adopted in positive and negative ion scanning with multiple reaction monitoring mode.RESULTS Seventeen constituents showed good linear relationships within their own ranges(r>0.9900),whose average recoveries were 87.2%-104.9%with the RSDs of 0.76%-6.55%.CONCLUSION This simple,rapid,sensitive and specific method can be used for the quality control of Buyang Huanwu Decoction.
