基于代谢组学技术解析精神分裂症患者血浆中代谢紊乱

Analyzing metabolic disorders in the plasma of schizophrenia patients based on metabolomics technology

摘要 目的:探求精神分裂症患者血浆中差异小分子代谢物表达特征及代谢通路,解析精神分裂症发生发展作用机制。方法:选取在本院就诊的精神分裂症患者30例为精神分裂症组,同期体检健康人群30名为健康对照组,采用代谢组学技术探究其血浆样本中小分子代谢物表达特征,以变量重要性投影值≥1及P<0.05为阈值,筛选精神分裂症患者差异表达代谢物,并对其进行代谢通路富集分析。结果:正、负两种离子模式下,分别鉴定了1432个和1036个代谢物,其中分别有163个和114个代谢物表达水平在两组间差异均具有统计学意义(P<0.05);差异代谢物主要为脂质、氨基酸、核苷酸等,富集于类固醇生物合成、嘧啶代谢、赖氨酸降解和脂肪酸生物合成等代谢通路。结论:精神分裂症患者血浆中小分子代谢物发生显著紊乱,涉及类固醇生物合成、嘧啶代谢和赖氨酸降解等代谢过程,与精神分裂症疾病进展息息相关。 Objective:To explore differential small-molecule metabolite expression profiles and metabolic pathways in the plasma of schizophrenia patients,and to elucidate their potential mechanisms in the pathogenesis and progression of schizophrenia.Method:Twenty-nine schizophrenia patients diagnosed and treated at our hospital were enrolled as the schizophrenia group,and thirty healthy individuals undergoing physical examinations were recruited as the healthy control group.Metabolomics technology was employed to investigate small-molecule metabolite expression profiles in plasma samples.Metabolites with variable importance in projection score≥1 and P<0.05 were screened as differentially expressed metabolites.Enrichment analysis was then performed to identify associated metabolic pathways.Results:Under positive and negative ion modes,1432 and 1036 metabolites were identified,respectively.Among these,163 and 114 metabolites showed statistically significant differential expression between the two groups(all P<0.05).The differentially expressed metabolites primarily included lipids,amino acids,and nucleotides,which were enriched in metabolic pathways such as steroid biosynthesis,pyrimidine metabolism,lysine degradation,and fatty acid biosynthesis.Conclusion:Significant disturbances in small-molecule metabolites were observed in the plasma of schizophrenia patients,involving key metabolic processes such as steroid biosynthesis,pyrimidine metabolism,and lysine degradation,which are closely associated with disease progression.
