摘要
目的:综合评价不同产地牛膝的质量,为经典名方中药材产地选择提供依据。方法:采集了河南、河北、内蒙古3个省区共计28批牛膝样品,测定其水分、总灰分、二氧化硫残留量、浸出物及β-蜕皮甾酮含量。采用化学计量学方法对牛膝的质量及分类情况进行评价。结果:样本的各项指标均符合《中国药典》(2020年版)规定,水分含量为4.80%~12.01%,总灰分含量为4.70%~6.81%,二氧化硫残留量为3.0~11.2 mg·kg^(-1),浸出物含量为6.71%~17.99%,β-蜕皮甾酮含量为0.0572%~0.1131%。依据主成分分析(PCA),河南孟州、温县、武陟所产牛膝的综合评分较高。聚类分析(CA)结果表明,所有样品分为2类,其中道地产区河南归为一类。正交偏最小二乘判别分析(OPLS-DA)结果表明,浸出物、总灰分、含水量的变量重要性投影值(VIP)大于1,表明产地的环境与加工方式可能是造成目前道地产区与非道地产区药材质量差异的主要因素。结论:不同产地间牛膝质量存在差异,其中河南产牛膝质量变异较小,总体质量优于其他产地。
Objective:To comprehensively evaluate the quality of Achyranthis Bidentatae Radix from different production areas and provide a reference for the selection of medicinal materials in classical famous formulas.Methods:A total of 28 batches of Achyranthis Bidentatae Radix samples were collected from three provinces/regions(Henan,Hebei,and Inner Mongolia).The moisture content,total ash,sulfur dioxide residue,extract content,and β-ecdysterone content were determined.Chemometric methods were used to evaluate the quality and classification of the samples.Results:All tested indicators of the samples complied with the standards of The Chinese Pharmacopoeia(2020 edition).The moisture content ranged from 4.80%to 12.01%,total ash from 4.70%to 6.81%,sulfur dioxide residue from 3.0 to 11.2 mg·kg^(-1),extract content from 6.71%to 17.99%,andβ-ecdysterone content from 0.0572%to 0.1131%.Based on principal component analysis(PCA),the samples from Mengzhou,Wenxian,and Wuzhi in Henan Province received higher comprehensive scores.Cluster analysis(CA)classified all samples into two groups,with the authentic producing area(Henan)forming a distinct cluster.Orthogonal partial least squares-discriminant analysis(OPLS-DA)revealed that extract content,total ash,and moisture content had variable importance in projection(VIP)values greater than 1,suggesting that environmental factors and processing methods may be the main contributors to the quality differences between authentic and non-authentic producing areas.Conclusion:There are significant differences in the quality of Achyranthis Bidentatae Radix from different origins.The samples from Henan exhibit smaller quality variations and generally superior quality compared to those from other regions.
