摘要
目前,多数高校的医学细胞生物学理论课程均进行了自噬相关内容讲解,但极少数高校开展自噬相关实验,教学效果欠佳。针对高校细胞生物学实验课内容与理论课知识明显脱节,滞后于学科发展现状,该研究基于小型模式生物使用设计了结构与功能相对应的自噬结构观察和功能探索的综合性教学实验,并招募基础医学、医学实验技术等专业学生开展开放性实验。该文以细胞自噬观察为例,探索利用基因修饰秀丽隐杆线虫为实验材料,通过梯度式实验设计以自噬诱导、自噬小体观察与统计、自噬对寿命影响等环节,有机连续地实施实验课程。教师课后通过小组文献调阅与讨论、撰写实验报告等环节,展示医学细胞生物学实验的课程理念,旨在通过教学改革实践探索,构建包含实验技能、文献研读、科学思维在内的科研能力培养体系,激发学生的科研热情,培养学生科研能力,丰富本科生细胞生物学实验教学内容,使学生更加深刻地理解细胞自噬对于生命演化的意义,为未来的科研工作奠定基础。
Currently,most medical schools teach autophagy-related content in the theoretical course of medical cell biology,yet very few succeed in carrying out autophagy experiments.There is an obvious disconnection between theoretical teaching and experimental teaching,which lags behind disciplinary development.Using genetically modified Caenorhabditis elegans as experimental material,the authors designed a comprehensive experiment to observe cellular autophagy and they recruited students of basic medicine and medical experimental technology to perform open experiments.The gradient-based experimental design features a series of modules of autophagy induction,autophagosome observation and quantification,and investigation of autophagy’s effects on lifespan.The authors also arranged after-class activities of group literature review,discussion,and experimental report writing.The authors established a cultivation system of scientific research ability,which is composed of experimental skills,literature reading,and scientific thinking.The teaching reform aims to increase students’enthusiasm,cultivate their research abilities,and enrich teaching content,so that the students can better understand autophagy’s significance in biological evolution,laying a solid foundation for their future scientific endeavors.
