摘要
研究测定了10批芍药(Paeonia lactiflora Pall.)茎和叶中芍药苷、没食子酸、苯甲酰芍药苷等主要成分的含量,对比芍药茎和叶中主要化学成分差异并通过抑菌实验对其抗菌活性进行评价。采用HPLC法,色谱条件:Agilent EC-C18柱(4.6 mm×150 mm,4 μm);流动相:乙腈-0.1%甲酸;流速:1.0 mL/min;检测波长:230 nm,梯度洗脱。UHPLC-MS/MS法,色谱条件:Waters-HSS T3柱(2.1 mm×100 mm,1.8 μm);流动相:乙腈-0.1%甲酸;流速:0.3 mL/min;检测波长:230 nm,梯度洗脱。结果显示芍药茎和叶均具有抗菌活性。芍药茎对gfp-E.coli和金葡菌的最小抑菌浓度分别为62.5、125 mg/mL;芍药叶对gfp-E.coli和金葡菌的最小抑菌浓度为62.5 mg/mL。本研究为综合评价芍药茎叶的质量和作为抑菌性食品添加剂提供相关理论基础。
The content of main components such as paeoniflorin,gallic acid,and benzoylpaeoniflorin in Paeonia lactiflora Pall.were determined in ten batches,differences in main chemical components in stem and leaf of Paeonia lactiflora Pall.were compared and the antibacterial activity was evaluated.HPLC analysis was performed under the following chromatographic conditions:Agilent EC-C18 column(4.6 mm×150 mm,4 μm);mobile phase:acetonitrile-0.1%formic acid;Flow rate:1.0 mL/min;Detection wavelength:230 nm,gradient elution was applied.UHPLC-MS/MS analysis was carried out using a Waters-HSS T3 column(2.1 mm×100 mm,1.8 μm);mobile phase:acetonitrile-0.1%formic acid;flow rate:0.3 mL/min;detection wavelength:230 nm,gradient elution was applied.It was found that both stem and leaf exhibited antimicrobial activity.The MIC of stem against gfplabeled E.coli and Staphylococcus aureus were determined to be 62.5 and 125 mg/mL respectively;the MIC of leaf against gfp-labelled E.coli and Staphylococcus aureus were 62.5 mg/mL.This study provides a theoretical basis for the comprehensive evaluation of the quality of P.lactiflora stems and leaves and for their use as antibacterial food additives.
