摘要
以十六醇为起始原料,与环氧卤代烷和乙醇胺分别发生取代、脱水缩合反应得仲胺中间体;中间体与不同链长的直链脂肪酸甲酯经胺解反应,全合成类神经酰胺(Cer X1、Cer X2、Cer X3、Cer X4、Cer E),总收率均在70%以上,其结构经^(1)HNMR、^(13)CNMR和HRMS谱确证,纯度经HPLC检测为98.00%以上。该合成路线简短、原料价廉易得、反应条件安全、工艺简单、收率较高,且适用于工业级类神经酰胺的制备。通过考察在配方应用中其外观、肤感、晶型及稳定性等的差异性,阐明了不同链长对类神经酰胺应用稳定性的影响。实验结果表明Cer E具有良好的应用稳定性,十六碳链长具有最佳的配方适应性。
Starting from hexadecanol,a secondary amine intermediate was synthesized via substitution and dehydration condensation reactions with epoxyhaloalkanes and ethanolamine.Subsequent amidolysis with straight-chain fatty acid methyl esters of varying chain lengths produced ceramide analogues(Cer X1,Cer X2,Cer X3,Cer X4,Cer E)with overall yields exceeding 70%.Structural confirmation was achieved using^(1)HNMR,^(13)CNMR,and HRMS,while HPLC analysis demonstrated purities above 98.00%.This synthetic route is concise,employs inexpensive and readily available raw materials,operates under mild and safe conditions,and is suitable for industrial-scale production due to its simplicity and high yield.Evaluations of appearance,skin feel,crystal form,and formulation stability revealed that carbon chain length significantly influences application performance.Among the analogues,Cer E,with a hexadecyl chain,exhibited superior formulation stability and optimal adaptability for practical applications.
