摘要
目的研究邓老御清膏(桑叶、枸杞子、菊花、决明子、山楂、荷叶)对高脂乳剂诱导高血脂症大鼠的辅助降血脂作用,并探究其潜在作用机制。方法本研究采用SD大鼠作为实验对象,取血清及肝脏测定各组大鼠血清总胆固醇(total cholesterol,TC)、甘油三酯(triglycerides,TG)、低密度脂蛋白(low-density lipoprotein cholesterol,LDL-C)、高密度脂蛋白(high-density lipoprotein cholesterol,HDL-C)、同型半胱氨酸(homocysteine,Hcy)、载脂蛋白A(apolipoprotein A,apo-A)、白细胞介素-6(interleukin-6,IL-6)、氧化低密度脂蛋白(oxidized low-density lipoprotein,Ox-LDL)、超氧化物歧化酶(superoxide dismutase,SOD)活性、丙二醛(malondialdehyde,MDA)含量及一氧化氮(nitrogen monoxide,NO)水平;观察大鼠体重、肝脏重量系数以及肝脏和主动脉组织病理学变化,并作出相关评价。结果与模型对照组比较,给药第3周,邓老御清膏中、高剂量组大鼠体重降低约18.0%,降幅显著(P<0.05);血清TC、TG、LDL-C、IL-6与MDA含量显著均降低(P<0.05),Hcy与Ox-LDL含量也降低,两者无统计学差异(P>0.05);但显著提高了HDL-C水平和SOD活性(P<0.05),以及apo-A和NO水平,两者无统计学差异(P>0.05);大鼠肝脏重量系数显著降低(P<0.05),脂肪颗粒明显减少,肝细胞形态保持正常,排列较为规整,脂肪变性较轻,各给药肝脏病理评分显著降低(P<0.05)。结论邓老御清膏能够调节高脂血症大鼠的血脂水平,有效保护肝组织,可能与调节机体脂质代谢、抗炎与抗氧化机制有关,为功能性食品药食同源组分可能有助于早期干预高脂血症高危人群提供了理论依据。
Objective To study the auxiliary lipid-lowering effect of TCM Master DENG Tietao’s Yu Qing Soft Extract(mulberry leaf,wolfberry,chrysanthemum,cassia seed,hawthorn,lotus leaf)on hyperlipidemia rats induced by high-fat emulsion,and to explore its potential mechanism of action.Methods In this study,SD rats were used as the experimental subjects,and serum and liver were taken to determine the activity of total cholesterol(TC),triglycerides(TG),low-density lipoprotein(LDL‑C),high-density lipoprotein(HDL‑C),homocysteine(Hcy),apolipoprotein A(apo-A),interleukin-6(IL-6),oxidized low-density lipoprotein(Ox-LDL),superoxide dismutase(SOD),malondialdehyde(MDA)content and nitrogen monoxide(NO)levels in each group of mice.The body weight,liver weight coefficient and histopathological changes in the liver and aorta were observed,and relevant evaluations were made.Results Compared with the rats in the model group,after the 3rd week of administration,the body weight of rats in the medium and high dose groups of the TCM Master DENG Tietao’s Yu Qing Soft Extract decreased by about 18.0%,a significant decrease(P<0.05);the serum TC,TG,LDL-C,IL-6 and MDA content significantly decreased(P<0.05).Hcy and Ox-LDL content also decreased,with no statistical difference between them(P>0.05);but HDL-C level and SOD activity significantly increased(P<0.05),as well as apo-A and NO levels,with no statistical difference between the two(P>0.05).The liver weight coefficient of the rats significantly decreased(P<0.05).Fat granules significantly decreased,the morphology of hepatocytes was still neat and the arrangement was relatively regular,and steatosis was not obvious,the pathological scores of the livers administered in each group were significantly lower(P<0.05).Conclusion The TCM Master DENG Tietao’s Yu Qing Soft Extract can regulate the blood lipid levels of rats with hyperlipidemia and effectively protect liver tissue,which may be related to the regulation of lipid metabolism in the body,anti-inflammatory and antioxidant mechanisms.It provides a theoreticalbasis for homology of medicine and food components in functional foods that may help early intervention in high-risk groups in hyperlipidemia.
