摘要
试验旨在探究槲皮素对脂多糖(LPS)诱导的肉鸡肠炎免疫保护作用的影响。选取300只1日龄体况良好的爱拔益加(AA)肉鸡,随机分为5组:低剂量组(基础饲粮+200 mg/kg槲皮素)、中剂量组(基础饲粮+400 mg/kg槲皮素)、高剂量组(基础饲粮+800 mg/kg槲皮素)、LPS组(基础饲粮)和对照组(基础饲粮),每组6个重复,每个重复10只鸡。试验期21 d。试验第16、18和20天,对低、中、高剂量组及LPS组肉鸡注射500μg/kg LPS溶液,对照组注射等量生理盐水。结果显示,中、高剂量组空肠绒毛高度和隐窝深度显著高于低剂量组和LPS组(P<0.05)。中剂量组法氏囊指数显著低于低、高剂量组(P<0.05)。中剂量组回肠白细胞介素-1β(IL-1β)、白细胞介素-8(IL-8)和干扰素-γ(IFN-γ)表达水平均显著低于低、高剂量组(P<0.05);中、高剂量组肿瘤坏死因子-α(TNF-α)表达水平均显著低于低剂量组(P<0.05)。中剂量组空肠TNF-α表达水平显著低于低、高剂量组(P<0.05)。中剂量组回肠封闭蛋白-2(Claudin-2)和空肠闭锁小带蛋白-2(ZO-2)表达水平均显著高于低、高剂量组(P<0.05)。研究表明,饲粮中添加400 mg/kg槲皮素可有效缓解LPS诱导的肉鸡肠道炎症,增强肠道屏障功能,改善肠道形态结构,并调节免疫反应。
The experiment aimed to investigate the effects of quercetin on immune protection against lipopolysaccharide(LPS)-induced enteritis in broilers.A total of 300 one-day-old Arbor Acres(AA)broilers in good condition were randomly divided into five groups:Low-dose group(basal diet+200 mg/kg quercetin),medium-dose group(basal diet+400 mg/kg quercetin),high-dose group(basal diet+800 mg/kg quercetin),LPS group(basal diet),and control group(basal diet).Each group contained six replicates with ten broilers per replicate.The test period was 21 days.On days 16,18,and 20 of the experiment,broilers in the low-dose,medium-dose,and high-dose groups as well as the LPS group were injected with 500μg/kg LPS solution,while the control group received an equal volume of physiological saline.The results showed that the jejunal villus height and crypt depth in the medium-dose and high-dose groups were significantly higher than those in the low-dose and LPS groups(P<0.05).The bursa of Fabricius index in the medium-dose group was significantly lower than that in the low-dose and high-dose groups(P<0.05).The expression levels of interleukin-1β(IL-1β),interleukin-8(IL-8),and interferon-γ(IFN-γ)in the ileum of the medium-dose group were significantly lower than those in the low-dose and high-dose groups(P<0.05).The expression level of tumor necrosis factor-α(TNF-α)in the medium-dose and high-dose groups was significantly lower than in the low-dose group(P<0.05).The TNF-αexpression level in the jejunum of the medium-dose group was significantly lower than that in the low-dose and highdose groups(P<0.05).The expression levels of the ileum Claudin-2 and the jejunum zonula occludens-2(ZO-2)in the medium-dose group were significantly higher than those in the low-dose and high-dose groups(P<0.05).The study indicates that dietary supplementation with 400 mg/kg quercetin can effectively alleviate LPS-induced intestinal inflammation in broilers,enhance intestinal barrier function,improve intestinal morphological structure,and modulate immune responses.
