OPA1介导线粒体动力学与心力衰竭的作用及中药调控研究进展

摘要 心力衰竭(HF)是一种多因素驱动的终末期心血管疾病,其核心特征为心肌细胞结构重塑、能量代谢障碍与细胞凋亡加剧。视神经萎缩蛋白1(OPA1)是一种定位于线粒体内膜的关键GTP酶,主要通过调控线粒体内膜融合与嵴结构稳定,维持线粒体网络的完整性、电化学膜电位、呼吸链功能及ATP生成。HF的核心病理过程涉及OPA1功能障碍驱动的线粒体过度碎片化,导致心肌能量崩溃与细胞死亡。本文从OPA1介导的线粒体动力学与HF的发病机制入手,系统分析中药单味及其活性成分与中药复方通过多靶点调控OPA1稳定线粒体动力学防治HF的机制,以期为HF的线粒体靶向治疗提供新的机制基础及潜在药物策略。 Heart failure(HF)is a multifactor-driven end-stage cardiovascular disease characterized by cardiomyocyte structural remodeling,energy metabolism disorders,and increased apoptosis.Optic atrophy 1(OPA1),a key GTP ase localized to the mitochondrial inner membrane,plays a vital role in maintaining mitochondrial network integrity by regulating inner membrane fusion and cristae structural stability.This regulation ensures the preservation of mitochondrial membrane potential,respiratory chain activity,and ATP production.A central pathological feature of HF involves excessive mitochondrial fragmentation driven by OPA1 dysfunction,leading to myocardial energy collapse and cell death.Starting from the pathogenesis of OPA1-mediated mitochondrial dynamics and HF,this paper systematically analyzes the mechanism of traditional Chinese medicine single flavor and its active ingredients and traditional Chinese medicine compound in preventing and treating heart failure by regulating OPA1 to stabilize mitochondrial dynamics through multi-target regulation,in order to provide a new mechanism basis and potential drug strategy for mitochondrial targeted therapy for HF.
