诺丽精油O/W型微乳的制备及其性能和抑菌活性研究

摘要 为了提高诺丽精油(Noni Essential Oil,NEO)的稳定性,将其进行微乳包埋,制备O/W型诺丽精油微乳(NEO Microemulsion,NEOM)。本研究以NEO为油相,以蓖麻油聚氧乙烯醚40(EL-40)为表面活性剂,采用水滴定法制备NEOM,通过拟三元相图计算微乳区面积结合微乳表观变化,考察助表面活性剂、Km值(表面活性剂与助表面活性剂的质量比)、组分及比例对微乳液形成的影响,并测定NEOM的粒径、构型、电导率、水溶性、稳定性及抑菌活性。结果表明,EL-40为表面活性剂,丙二醇为助表面活性剂,Km为3时,NEO与混合表面活性剂质量比为7:3,O/W型水相的含水量为70%时,可以形成的微乳稳定、透明澄清,且平均粒径在(25.53±3.09) nm,PDI为0.180±0.03的O/W型微乳液。同时,NEOM具有良好的贮藏稳定性、离心稳定性和盐离子浓度稳定性,适宜在偏酸性环境下以及45℃以下使用。NEOM对白色念珠菌的抑菌作用优于NEO。 In order to improve the stability of Noni Essential Oil(NEO),a stable O/W type NEO Microemulsion(NEOM)was prepared by embedding it in microemulsion.In this study,NEO was used as the oil phase,and castor oil polyoxyethylene ether 40(EL-40)was used as the surfactant.NEOM was prepared by water titration method.The microemulsion area which calculated by drawing a pseudo-ternary phase diagram combined with the apparent change of microemulsion to investigate the effect of cosurfactant,Km value(mass ratio of surfactant to cosurfactant),composition and proportion on the formation of microemulsion.Then the particle size,configuration,conductivity,water solubility,stability and antibacterial activity of NEOM were further determined.The results showed that when EL-40 as the surfactant,propylene glycol as the cosurfactant,Km was 3 when the mass ration of NEO to surfactant was 7:3,and the water content of O/W phase was 70%,the NEOM could be formed with stable,transparent and clear appearance.The average particle size was(25.53±3.09)nm and the PDI was 0.180±0.03 of the 0/W NOEM.At the same time,NEOM had good storage stability,centrifugal stability and salt ion concentration stability,and is suitable for use in acidic environments and below 45℃.The antifungal activity of NEOM against Candida albicans was superior to NEO.
