元宝枫籽油纳米结构脂质载体对秀丽线虫衰老的改善作用

Antiaging Effects of Acer truncatum Seed Oil-based Nanostructured Lipid Carriers on Caenorhabditis elegan

摘要 该文对熔融乳化-超声法制备得到的元宝枫籽油纳米结构脂质载体(ASO-NLC)性状进行表征,并考察其对秀丽隐杆线虫寿命、生育能力、运动能力、抗氧化能力、热/氧化应激及细胞凋亡的影响。结果表明,ASO-NLC粒径为244.40 nm,PDI为0.17,zeta电位为-30.07 mV,离心稳定性好,是一种稳定性良好的口服制剂。与空白组比较,ASO-NLC可使线虫最长寿命延长50.21%以上,平均寿命延长12.00%~36.00%,而且寿命的延长不会导致线虫生殖能力下降;线虫培养10 d后,ASO-NLC能显著提高线虫的蠕动能力,到15 d时,作用更为明显。ASO-NLC可使谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)、超氧化物歧化酶(SOD)等抗氧化酶活性分别提高21.58%~48.81%、22.42%~58.58%和16.11%~75.88%;丙二醛(MDA)含量降低6.81%~34.89%。急性氧化应激下,ASO-NLC组线虫最长存活时间延长了1.50倍。吖啶橙染色结果显示,ASO-NLC组相对荧光强度比空白组减弱了13.71%~17.00%。综上所述,ASO-NLC可通过延长线虫寿命,提升其运动能力,提高线虫抗应激损伤的能力、体内抗氧化酶活性及减少细胞凋亡来延缓线虫衰老。该研究为元宝枫籽油功能性多元化产品开发提供了理论基础。 The properties of Acer truncatum seed oil-based nanostructured lipid carriers(ASO-NLC)prepared using the melt emulsification–ultrasonication method were characterized,and their effects on the longevity,fertility,exercise capacity,antioxidant capacity,heat/oxidative stress,and apoptosis of Caenorhabditis elegans were evaluated.ASO-NLC exhibited a particle size of 244.40 nm,a polydispersity index of 0.17,a zeta potential of−30.07 mV,and good centrifugal stability,that it is a stable oral formulation.Comparisons between the treatment groups and the control group(NC)revealed that ASONLC could extend the longest lifespan by more than 50.21%and the average lifespan by 12%~36%.Moreover,the extension of life span did not reduce the fertility of C.elegans.ASO-NLC also significantly improved the motility of C.elegans after 10 days of culture,with more obvious enhancement observed on day 15.Additionally,ASO-NLC increased the activities of glutathione peroxidase,catalase,and superoxide dismutase by 21.58%~48.81%,22.42%~58.58%,and 16.11%~75.88%,respectively,while reducing the malondialdehyde content by 6.81%~34.89%.Under acute oxidative stress,the longest survival time of C.elegans in the ASO-NLC group was 1.50-fold longer than that in the NC group.The acridine orange staining results demonstrated that the relative fluorescence intensity of the ASO-NLC group was 13.71%~17.00%,which was significantly lower than that of the NC group(P<0.01).In conclusion,ASO-NLC can increase the lifespan,exercise capacity,and stress resistance of C.elegans;enhance the in vivo activities of antioxidant enzymes;and reduce apoptosis,enabling ASO-NLC to slow the aging of C.elegans.This study provides a theoretical basis for the development of functional products of A.truncatum seed oil.
