基于通宣理肺方加减对风寒表证大鼠的影响分析麻黄升浮药性

Evaluation of Ascending and Floating Medicinal Properties of Ephedrae Herba Based on the Effects of Modified Tongxuan Lifei Formula in a Rat Model of Wind-cold Exterior Syndrome

摘要 目的:基于通宣理肺方加减对风寒表证大鼠的影响分析麻黄升浮药性。方法:80只SD雄性大鼠,采用编号随机法挑选10只为对照组,剩余构建风寒表证动物模型后,将大鼠分为模型组、对乙酰氨基酚组、麻黄组、生物碱组、通宣理肺方组、通宣理肺方减麻黄组和通宣理肺方减麻黄配伍生物碱组,给药5 d后,检测5大系统指标、大鼠肺组织中丝裂原活化蛋白激酶(MAPK)信号通路及皮肤环磷酸腺苷-蛋白激酶A/水通道蛋白5(cAMP-PKA/AQP5)信号通路相关蛋白表达。结果:与模型组比较,各给药组可改善呼吸系统功能,减轻肺组织病理损伤,降低肺泡灌洗液中白细胞介素4(IL-4)、免疫球蛋白(IgE)水平并升高γ干扰素(IFN-γ)水平;兴奋中枢系统,增加活动距离和次数;调节泌尿系统,增加汗尿量;对循环和消化系统无明显作用。此外,该方可明显降低肺组织中磷酸化-细胞外信号调节激酶1/2(p-ERK1/2)和磷酸化-丝裂原活化蛋白激酶(p-p38MAPK)的表达水平(P<0.01),增加腋下皮肤中AQP5、p-PKA、cAMP蛋白表达水平(P<0.01),减麻黄后药效减弱,配伍生物碱后各指标恢复至原方水平(P<0.01)。结论:通宣理肺方通过调节MAPK和cAMP-PKA/AQP5通路改善风寒表证,麻黄生物碱作为关键“升浮组分”,承担“宣发”“发散”核心药效,证实麻黄升浮药性的科学内涵。 Objective:To analyze the ascending and floating medicinal properties of Ephedrae Herba based on the effects of modified Tongxuan Lifei Formula on rats with wind-cold exterior syndrome.Methods:Eighty male SD rats were randomly assigned,with 10 selected as the control group.The remaining rats were used to establish a wind-cold exterior syndrome model and were divided into model group,acetaminophen group,Ephedrae Herba group,alkaloids group,Tongxuan Lifei Formula group,Tongxuan Lifei Formula minus Ephedrae Herba group,and Tongxuan Lifei Formula minus Ephedrae Herba combined with alkaloids group.After 5 days of drug administration,indicators of five major physiological systems were measured,along with the expression of MAPK pathway-related proteins in lung tissue and cAMP-PKA/AQP5 signaling pathway proteins in skin tissue.Results:Compared with the model group,all medication groups improved respiratory function,reduced pathological damage in lung tissue,decreased interleukin-4(IL-4)and immunoglobulin E(IgE)levels in bronchoalveolar lavage fluid,and increased interferon-γ(IFN-γ)levels.They also stimulated the central nervous system,increasing movement distance and frequency,and regulated the urinary system by increasing sweat and urine output.No significant effects were observed on the circulatory and digestive systems.In addition,the formula significantly reduced the expression levels of phosphorylated extracellular signal-regulated kinase 1/2(p-ERK1/2)and phosphorylated p38 MAPK(p-p38MAPK)in lung tissue(P<0.01),and increased the expression of AQP5,p-PKA,and cAMP in axillary skin(P<0.01).When Ephedrae Herba was removed,the efficacy weakened,but after combining with alkaloids,the indicators returned to levels similar to the original formula(P<0.01).Conclusion:Tongxuan Lifei Formula alleviates wind-cold exterior syndrome by modulating the MAPK and cAMP-PKA/AQP5 signaling pathways.Ephedrae Herba alkaloids act as key“ascending and floating”components,fulfilling the core functions of dispersing and ventilating,confirming the scientific basis of ascending and floating medicinal properties of Ephedrae Herba.
