基于薄层色谱法和高效液相色谱法的清热祛湿颗粒质量标准提升研究

摘要 目的基于薄层色谱法和高效液相色谱法,分别建立定性鉴别和含量测定方法,为清热祛湿颗粒质量标准提升提供参考依据。方法采用薄层色谱法对清热祛湿颗粒中的绿原酸、蒙花苷进行定性鉴别;以绿原酸、蒙花苷为对照品,通过高效液相色谱法对处方中的主要药味野菊花、岗梅根、茵陈进行含量测定。结果在采用的薄层色谱条件下,绿原酸、蒙花苷色谱斑点清楚,分离效果好,无干扰;在高效液相色谱含量测定中,绿原酸、蒙花苷均具有良好的线性关系,该方法绿原酸、蒙花苷的回收率分别为100.47%、99.63%。结论建立的定性鉴别和含量测定方法比较简便、准确、灵敏、重现性好,可用于清热祛湿颗粒质量标准提升。 Objective To establish qualitative identification and content determination methods based on thin-layer chromatography(TLC)and high-performance liquid chromatography(HPLC)to provide reference for the improvement of the quality standard of Qingre Qushi Granules.Methods TLC was used to qualitatively identify chlorogenic acid and luteolin-7-O-glucoside in Qingre Qushi keli.With chlorogenic acid and luteolin-7-O-glucoside as reference substances,HPLC was employed to determine the content of the main medicinal ingredients,including Chrysanthemum indicum L.,Ilex asprella(Hook.et Am.)Champ.ex Benth.and Artemisia capillaris Thunb.,in the prescription.Results Under the TLC conditions used,the chromatographic spots of chlorogenic acid and luteolin-7-O-glucoside were clear,with good separation and no interference.In the HPLC content determination,both chlorogenic acid and luteolin-7-O-glucoside exhibited good linear relationships.The recovery rates of chlorogenic acid and luteolin-7-O-glucoside using this method are 100.47%and 99.63%.Conclusion The established qualitative identification and content determination methods are simple,accurate,sensitive,and reproducible,and can be used for the improvement of the quality standard of Qingre Qushi Granules.
