摘要
目的:探讨乌头汤调控CircFOXO3改善由白细胞介素-1β(IL-1β)诱导的软骨细胞炎症的糖酵解的作用效果,为乌头汤的临床应用提供实验依据。方法:3周龄SPF级C57BL/6雄性小鼠20只,提取小鼠双膝关节软骨进行体外软骨细胞的分离与培养,用10 ng/mLIL-1β作用24 h诱导体外软骨细胞模型,构建CircFOXO3过表达细胞株;将软骨细胞随机分为空白组、IL-1β组、乌头汤组(10 ng/mL IL-1β+10%乌头汤含药血清)、oe-CircFOXO3组和oe-CircFOXO3+乌头汤组。通过Real-time PCR检测各组软骨细胞中CircFOXO3的相对表达水平,通过Western blot检测各组软骨细胞中GLUT1、PKM2、HK2和LDHA的蛋白含量。结果:CircFOXO3在IL-1β诱导的软骨细胞模型中表达显著降低(P<0.05);与空白组相比,IL-1β组中GLUT1、PKM2、HK2和LDHA蛋白表达增强(P<0.05);与oe-CircFOXO3组比较,oe-CircFOXO3+乌头汤组上述蛋白含量下降(P<0.05);与IL-1β组相比,乌头汤组中CircFOXO3表达增强(P<0.05)。结论:乌头汤可通过调控CircFOXO3抑制骨关节炎软骨细胞糖酵解,改善软骨退变。
Objective:To explore the effect and mechanism of auriculotherapy in modulating CircFOXO3 to improve glycolysis in chondrocyte inflammation induced by interleukin-1β(IL-1β),and to provide experimental basis for the clinical application of auriculotherapy.Methods:203-week-old SPF C57BL/6 male mice were extracted from the cartilage of both knee joints for the isolation and culture of chondrocytes in vitro,and the chondrocyte model was induced by the action of 10 ng/mL IL-1βfor 24 h in vitro,and CircFOXO3 overexpression cell line was constructed;chondrocytes were randomly divided into a blank group,an IL-1βgroup,an aconite soup group(10 ng/mL IL-1β+10%Wutou decoction containing serum),oe-CircFOXO3 group and oe-CircFOXO3+Wutou decoction group.The relative expression level of CircFOXO3 in chondrocytes of each group was detected by Real-time PCR.Western blot detected the protein content of GLUT1,PKM2,HK2 and LDHA in chondrocytes of each group.Results:CircFOXO3 expression was significantly reduced in the IL-1β-induced chondrocyte model(P<0.05),and the protein expression of GLUT1,PKM2,HK2 and LDHA was enhanced in the IL-1βgroup compared with the blank group(P<0.05);the above protein levels were decreased in the oe-CircFOXO3+Wutou decoction group compared with the oe-CircFOXO3 group(P<0.05),and the protein levels of the above mentioned proteins were decreased in the oe-CircFOXO3+Wutou decoction group compared with the IL-1βgroup,CircFOXO3 expression was enhanced in the oe-CircFOXO3 group(P<0.05).Conclusion:Wutou Decoction can inhibit osteoarthritic chondrocyte glycolysis and improve cartilage degeneration by regulating CircFOXO3.
