离子色谱法与紫外分光光度法测定尿液中亚硝酸盐的方法学比较

摘要 比较和分析离子色谱法和紫外分光光度法测定尿液中亚硝酸盐方法的异同。对这两种方法的检出限、线性范围、精密度、回收率进行对比。离子色谱法的线性范围在0.00~2.00 mg/L,其线性关系较好;紫外分光光度法的线性范围在0.00~0.50 mg/L,其线性关系较好,相关系数R2均大于0.999。离子色谱法测得的亚硝酸盐的加标回收率为89.3%~97.4%,相对标准偏差(RSD)为0.96%~5.37%;紫外分光光度法测得的亚硝酸盐的加标回收率为92.4%~101%,相对标准偏差(RSD)为1.03%~4.26%。离子色谱法与紫外分光光度法均具有较好的检出限、线性范围、精密度以及准确度。尿液中,两种方法的检验结果无显著性差异,离子色谱法选择性好、操作简便、自动化程度高;紫外分光光度法分析时间短,成本低。两种方法均可用于尿液中亚硝酸盐的准确测定。 To Comparison and analysis of the ion chromatography and ultraviolet spectrophotometry method for the determi⁃nation of nitrite in urine.The detection limit,linear range,precision,and recovery rate of the two methods for the determina⁃tion of nitrite content in urine were compared.The linear range of nitrite measured by ion chromatography was between 0.00-2.00 mg/L,and the linear range nitrite measured by ultraviolet spectrophotometry was between 0.00-0.50 mg/L,both with good linear relationship and correlation coefficient R2 greater than 0.999.The recovery rate of nitrite measured by ion chromatography was 89.3%-97.4%,with a relative standard deviation(RSD)of 0.96%-5.37%;the recovery rate of nit mea⁃sured by ultraviolet spectrophotometry was 92.4%-101%,with a relative standard deviation(RSD)of 1.03%-4.26%.Conclu⁃sion Both ion chromatography and ultraviolet spectrophotometry have good detection limits,linear ranges,precision,accura⁃cy.There was no significant difference in the test results of the two methods in urine,and ion chromatography has good se⁃lectivity,simple operation,and high degree automation;ultraviolet spectrophotometry has short analysis time and low cost.Both methods can be used for the accurate determination of nitrite in urine.
