金耳多糖理化分析及其降血糖活性潜力研究 被引量:1

摘要 以甘肃省兰州地区培育的金耳子实体为原料,经分离纯化得到分子质量(M_(w)=1.54×10^(6)Da)分布均一的金耳纯多糖(Tremella aurantialba pure polysaccharide,TAPP),其主要单糖组成为甘露糖∶葡萄糖醛酸∶葡萄糖∶木糖=55.20∶9.76∶14.81∶20.23,含有少量半乳糖和半乳糖醛酸。傅里叶变换红外光谱、刚果红实验、扫描电子显微镜分析和热重分析结果表明,TAPP含有β-糖苷键和螺旋结构,表面微观形貌呈片状无规则聚集状态,170℃以下热稳定性良好。活性分析结果显示,TAPP对α-淀粉酶的和α-葡萄糖苷酶的抑制与浓度呈正相关,其对α-葡萄糖苷酶的抑制效果尤为突出,TAPP浓度为2.0 mg/mL时,对α-葡萄糖苷酶的抑制率为(93.32±1.09)%,与阿卡波糖效果相近(P<0.05);TAPP给药浓度达到75μg/mL时,可以显著改善棕榈酸诱导的HepG2-IR模型细胞的胰岛素抵抗症状(P<0.05)。该研究结果为兰州地区金耳资源的充分开发和高值化利用提供了一定理论支持和开发基础。 Fruiting body of Tremella aurantialba cultivated in Lanzhou was used for experimental material.Tremella aurantialba pure polysaccharide(TAPP),an acidic heteropolysaccharide,was obtained from the material,with uniform molecular weight distribution(M_(w)=1.54×10^(6)Da).Monosaccharide composition indicated that TAPP was composed of Man,GlcA,Glc,and Xyl with the ratio of 55.20∶9.76∶14.81∶20.23,and included a little Gal and GalA.The experiences of Fourier transform infrared spectroscopy,Congo red,scanning electron microscope,and thermogravimetric analysis showed that TAPP containedβ-glycosidic bonds and helical structure,surface micromorphology presented lamellar and irregular aggregation,and it had good chemical stability below 170℃.The activity analysis showed that the inhibition of TAPP onα-amylase andα-glucosidase was positively correlated with concentration.The inhibitory effect onα-glucosidase was more significant,and the inhibition rate reached(93.32±1.09)%at TAPP concentration of 2.0 mg/mL.Its inhibition rate was similar to the effect of acarbose(P<0.05).It could significantly improve the insulin resistance of HepG2-IR model cells induced by palmitic acid when the concentration of TAPP reached 75μg/mL(P<0.05).The results of this study provide theoretical supports and basis for further development and utilization of Tremella aurantialba resources in Lanzhou.
