响应面法优化刺梨总黄酮提取工艺及其抗氧化活性研究

Study on optimization of the extraction process and antioxidant activity of total flavonoids from Rosa roxbunghii by response surface method

摘要 试验旨在优化刺梨总黄酮提取工艺并测定其抗氧化活性。试验采用响应面法优化超声波辅助提取刺梨总黄酮提取工艺参数,并测定刺梨总黄酮的抗氧化活性。结果显示,刺梨总黄酮的最优提取工艺参数为:乙醇浓度为70%、浸提温度为60℃、浸提时间为45 min,重复3次试验,在最优条件下测得刺梨总黄酮提取得率为4.86%,与预测值基本一致,表明该方法可准确模拟和预测黄酮得率。刺梨总黄酮对1,1-二苯基-2-三硝基苯肼(DPPH)、2,2-联氮-二(3-乙基-苯并噻唑啉-6-磺酸)二铵盐(ABTS)自由基的IC_(50)值分别为235.6 mg/L、306.8 mg/L。研究表明,刺梨总黄酮采用响应面法优化超声波辅助提取工艺方案可行,且刺梨总黄酮具有较好的抗氧化功能。 The experiment aimed to optimize the extraction process of total flavonoids from Rosa roxbunghii and determine its antioxidant activity.The response surface methodology was used to optimize the ultrasonic-assisted extraction process parameters of total flavonoids from Rosa roxbunghii,and the antioxidant activity of total flavonoids from Rosa roxbunghii was determined.The results showed that the optimal extraction process parameters of total flavonoids from Rosa roxbunghii were:Ethanol concentration of 70%,extraction temperature of 60℃,and extraction time of 45 min.Three repeated experiments were conducted,and the extraction yield of total flavonoids from Rosa roxbunghii was 4.86%under the optimal conditions,which was basically consistent with the predicted value,indicating that this method could accurately simulate and predict the flavonoid yield.The IC_(50)values of total flavonoids from Rosa roxbunghii against 1,1-diphenyl-2-picrylhydrazyl(DPPH)and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt(ABTS)free radicals were 235.6 and 306.8 mg/L,respectively.The study indicated that the ultrasonicassisted extraction process of total flavonoids from Rosa roxbunghii optimized by response surface methodology was feasible,and total flavonoids from Rosa roxbunghii had good antioxidant function.
