基于16S rDNA高通量测序技术探究三味阿拉坦其其格散对功能性消化不良大鼠肠道菌群的影响 被引量:1

Effects of Alatanqiqige-3 on intestinal flora of rats with functional dyspepsia based on 16S rDNA high-throughput sequencing

摘要 目的:研究三味阿拉坦其其格散对功能性消化不良(FD)大鼠肠道菌群的影响。方法:将SD大鼠32只随机分成正常组、模型组、多潘立酮组、三味阿拉坦其其格散组,采用不规则喂养加夹尾刺激法建立FD大鼠模型,造模同时药物干预15 d,观察各组大鼠一般状态,测定胃内残留率及小肠推进率,用16S rDNA高通量测序技术分析三味阿拉坦其其格散对FD大鼠肠道菌群的影响。结果:与正常组比较,模型组大鼠胃内残留率显著上升(P<0.05),小肠推进率显著降低(P<0.05)。与模型组比较,给药组大鼠胃内残留率显著降低(P<0.05),小肠推进率显著上升(P<0.05)。FD大鼠用三味阿拉坦其其格散干预后,大鼠肠道菌群结构有明显变化,与模型组比较,三味阿拉坦其其格散组显著提高了肠道菌群的丰富性和多样性(P<0.01,P<0.05)。与模型组比较,三味阿拉坦其其格散组厚壁菌门与拟杆菌门的比值明显提高,乳酸杆菌属的相对丰度明显增加。三味阿拉坦其其格散干预FD大鼠后改变了肠道菌群优势菌种相对丰度。结论:三味阿拉坦其其格散能够改善FD大鼠的症状,调节其肠道菌群结构,上调有益菌相对丰度,提高肠道菌群丰富性和多样性,从而成为防治FD的重要途径之一。 Objective:To study the effect of Alatanqiqige-3 on the intestinal flora of rats with functional dyspepsia(FD).Methods:Thirty-two SD rats were randomly divided into a normal group,model group,domperidone group and Alatanqiqige-3 group,and the FD rat model was established by iregular feeding plus tail-clamping stimulation method.Modeling and simultaneous drug intervention were conducted for 15 d.The general condition of the rats in each group was observed,and the rate of intragastric retention and the propulsion rate in the small intestine were determined,and the effect of Alatanqiqige-3 on the intestinal flora of FD rats was analyzed by 16S rDNA high-throughput sequencing technology.Results:Compared with the normal group,the rate of intragastric retention was significantly increased(P<0.05)and the rate of small intestinal propulsion was significantly decreased(P<0.05)in the model group of rats.Compared with the model group,the rate of intragastric retention was significantly decreased(P<0.05)and the rate of small intestinal propulsion was significantly increased(P<0.05)in the administered rats.After FD rats were intervened with Alatanqiqige-3,there were significantly altered the structure of the intestinal flora of the rats,and intervention with Alatanqiqige-3 group significantly increased the abundance and diversity of the intestinal flora when compared with the model group(P<0.01,P<0.05).Compared with the model group,the ratio of Firmicutes and Bacteroidets(F/B ratio)of Alatanqiqige-3 group was significantly increased and the relative abundance of Lactobacillus was significantly increased.Intervention with FD rats by Alatanqiqige-3 altered the relative abundance of dominant species of intestinal flora.Conclusion:The Alatanqiqige-3 can improve the symptoms of FD rats,regulate the intestinal flora structure of FD rats,up-regulate the abundance of probiotics,and improve the richness and diversity of intestinal flora.It may become one of the important ways to prevent and treat FD.
