摘要
目的研究健脾理气方对功能性消化不良(functional dyspepsia,FD)大鼠中枢神经系统和外周十二指肠促肾上腺皮质激素释放因子(corticotropin releasingfactor,CRF)及CRF受体1(CRF-R1)的分布与表达的影响,探讨健脾理气方对FD治疗作用的可能机制。方法36只雄性大鼠随机分为3组,并接受幼年碘乙酰胺灌胃联合夹尾应激刺激方法建立FD动物模型,其中正常组及模型组予生理盐水灌胃,治疗组予健脾理气方中药汤剂灌胃。分别采用Western Blot(WB)或免疫组化染色(IHC)分析下丘脑、脊髓及十二指肠中CRF的分布及表达,定量PCR(qPCR)检测十二指肠中CRF-R1的表达水平。结果与正常对照组相比,模型组大鼠下丘脑、脊髓及十二指肠中CRF表达水平均明显升高(P<0.01),十二指肠组织中CRF-R1 mRNA表达水平明显增加(P<0.01);健脾理气方治疗组以上各指标均较模型组显著降低(P<0.01,P<0.01,P<0.05,P<0.01)。结论健脾理气方可能通过调节中枢神经系统及外周十二指肠CRF及CRF-R1的表达,从而对FD起到治疗作用。
Objective To investigate the distribution and expression of corticotropin releasing factor(CRF)and CRF receptor1(CRF-R1)in central nervous system and/or peripheral duodenum of rats with functional dyspepsia(FD)treated by Jianpi Liqi Formula,and to explore the possible mechanism of Jianpi Liqi Formula in treating FD.Methods Thirty-six male rats were randomly divided into three groups.The model of FD was established by intragastric administration of iodoacetamide combined with tail-clamping stress stimulation.The normal group and model group were intragastrically administered with normal saline,while the treatment group was intragastrically administered with Jianpi Liqi Formula.The expression of CRF in hypothalamus,spinal cord and duodenum were analyzed by Western blot(WB)or immunohistochemical staining(IHC),and the expression of CRF-R1 in duodenum was detected by quantitative PCR(qPCR).Results Compared with the normal control group,the expression of CRF in hypothalamus,spinal cord and duodenum in model group was significantly increased(P<0.01),and the expression of CRF-R1 in duodenum was significantly increased(P<0.01);the above indexes in Jianpi Liqi Formula treatment group were significantly lower than those in the model group(P<0.01,P<0.01,P<0.05,P<0.01).Conclusion Jianpi Liqi Formula plays a therapeutic role in FD by regulating the expression of CRF and CRF-R1 in central nervous system and peripheral duodenum.
