产蛋白酶芽孢杆菌的筛选鉴定、生长特性及全基因组分析

摘要 本研究通过产蛋白酶培养基筛选具有产蛋白酶能力的芽孢杆菌并进行菌株鉴定;利用生长曲线、培养温度、pH及NaCl耐受性测定,研究该菌株的生长特性;采用全基因组分析挖掘其产蛋白基因。结果表明:筛选获得产蛋白酶能力的菌株FJAT-26384,该菌株的蛋白酶活性为66.67 U/mL。经鉴定,该菌株为侧孢短芽孢杆菌(Brevibacillus laterosporus)。该菌株在30~50℃,pH 7~9及NaCL浓度为0~2%时均可生长。菌株FJAT-26384的基因组由1条环状染色体组成,大小约为5523271 bp,GC含量为40.72%。经Antismash 5.1.2预测和分析,菌株FJAT-26384基因组具有19个次级代谢产物基因簇;通过对菌株FJAT-26384全基因组上蛋白酶相关基因的筛查与统计发现,共筛查到33个与蛋白酶相关的基因。 In this study,Bacillus with protease-producing capacity was screened by protease-producing medium and the strains were identified.Growth curve,culture temperature,pH and NaCl tolerance were used to study the growth characteristics of this strain.The protein-producing genes were extracted by whole genome analysis.The results showed that strain FJAT-26384 with protease production ability was selected,and its protease activity was 66.67 U/mL.The strain was identified as Brevibacillus laterosporus.The strain could grow at 30~50℃,pH 7~9 and NaCl concentration 0~2%.The genome of strain FJAT-26384 consisted of a ring chromosome with a size of 5523271 bp and a GC content of 40.72%.Predicted and analyzed by Antismash 5.1.2,the genome of strain FJAT-26384 had 19 secondary metabolite gene clusters.Through the screening and statistics of protease-related genes in the whole genome of strain FJAT-26384,a total of 33 protease-related genes were detected.
