摘要
【目的】从福建平潭盐碱地菌草中筛选耐盐碱内生细菌,明确其分类地位、耐盐碱能力、生理生化特性及对玉米的促生效果,为盐碱地改良及菌草应用提供理论支持。【方法】利用含1.0 mol/L NaCl的LB固体培养基筛选获得耐盐碱菌草内生菌株PXY-25和PXY-28,采用盐浓度梯度与pH梯度试验测定其耐盐碱能力,结合16Sr RNA序列分析进行分子鉴定;利用平板试验研究其生理生化特性及对4种病原真菌的拮抗能力;通过盆栽试验分析其对玉米地下部生长指标、抗氧化酶活性及叶绿素含量的影响。【结果】2株耐盐碱内生细菌均鉴定为路氏肠杆菌(Enterobacter ludwigii),菌株PXY-25可在1.5 mol/L NaCl、0.05 mol/L Na2CO3及pH为10.0的LB培养基中生长,菌株PXY-28可在1.5 mol/L NaCl、0.05 mol/L Na2CO3及pH为9.0的LB培养基中生长;2个菌株均能溶解无机磷、产生接触酶及产铁载体,对炭疽属菌(PHZ-8)有一定抑制作用。玉米盆栽试验结果表明,与对照相比,PXY-25处理玉米根长增加56.95%,PXY-28处理玉米根长增加19.79%。PXY-25和PXY-28处理均可降低玉米丙二醛(MDA)含量、分别比对照降低49.09%和60.92%,且PXY-25处理显著提高玉米总叶绿素含量、比对照增加94.78%。【结论】菌株PXY-25具有较强的耐盐碱能力和显著的玉米促生特性,可作为盐碱地改良及作物促生的潜在菌株。
【Objective】This study aims to isolate salt-alkali tolerant endophytic bacteria from JUNCAO in the saline-alkali soils of Pingtan,Fujian,clarify their taxonomic status,salt-alkali tolerance,physiological and biochemical characteristics,and their growth-promoting effects on maize,providing theoretical support for the improvement of saline-alkali land and the application of JUNCAO.【Method】The salt-tolerant bacterial strains PXY-25 and PXY-28 were obtained through screening with 1.0 mol/L NaCl.Their salt-alkali tolerance was determined using salt concentration gradients and pH gradients,combined with molecular identification through 16S rRNA sequence analysis.Plate assays were used to study their physiological and biochemical characteristics as well as their antagonistic abilities against four pathogenic fungi.Additionally,pot experiments were conducted to analyze their effects on corn underground growth indicators,antioxidant enzyme activity,and chlorophyll content.【Result】Two salt-alkali tolerant endophytic bacteria were identified as Enterobacter ludwigii.PXY-25 can grow in LB medium with 1.5 mol/L NaCl,0.05 mol/L Na2CO3,and a pH of 10.0,while PXY-28 can grow in LB medium with 1.5 mol/L NaCl,0.05 mol/L Na2CO3,and a pH of 9.0.Both strains can dissolve inorganic phosphorus,produce contact enzymes and weak iron carriers,and have a certain effect on the anthracnose bacteria(PHZ-8).In the corn pot experiment,PXY-25 significantly increased root length,with an increase of 56.95%compared to the control.The root length of corn treated with PXY-28 increased by 19.79%compared to the control.Both strains reduced the malondialdehyde(MDA)content in corn,decreasing it by 49.09%and 60.92%respectively compared to the control.Among them,PXY-25 significantly increased the total chlorophyll content,with an increase of 94.78%compared to the control.【Conclusion】PXY-25 has strong salt-alkali tolerance and significant growth-promoting characteristics for maize,making it a potential strain for the improvement of saline-alkali land and the promotion of crop growth.
