响应面法优化地黄块根总多酚提取工艺及其抗氧化活性研究

Study on optimization of total polyphenol extraction process from root of Rehmannia glutinosa by response surface method and its antioxidant activity

摘要 试验旨在采用响应面法优化地黄块根总多酚的提取工艺,并探究其体外抗氧化能力。采用超声辅助结合Folin-Ciocalteu比色法提取地黄块根中总多酚。选取液料比、乙醇质量分数、提取温度和提取时间作为试验因素,以总多酚提取率为指标,在单因素试验基础上,结合响应面法对其提取工艺进行优化,并测定地黄块根总多酚清除1,1-二苯基-2-三硝基苯肼(DPPH)自由基和羟基自由基的能力及其还原能力,以评价总多酚的体外抗氧化活性。结果显示,地黄块根总多酚的最佳提取工艺为液料比10 mL/g、乙醇质量分数83%、提取温度68℃、提取时间2.3 h,此时地黄块根总多酚提取率为16.131%。地黄块根总多酚对DPPH自由基和羟基自由基的最大清除率分别为85.94%和60.81%,且还原能力较好。研究表明,地黄块根总多酚的抗氧化活性较强,可作为天然绿色抗氧化剂在动物生产中应用。 The experiment aimed to optimize the extraction process of total polyphenols from the root of Rehmannia glutinosa using the response surface methodology,and to investigate their in vitro antioxidant capacity.The total polyphenols in the root of Rehmannia glutinosa were extracted using an ultrasonic-assisted method combined with the Folin-Ciocalteu colorimetric assay.The liquid-to-solid ratio,ethanol concentration,extraction temperature,and extraction time were selected as the experimental factors.The total polyphenol extraction rate was used as the indicator.Based on the single-factor experiments,the extraction process was optimized using the response surface methodology.The ability of total polyphenols from the root of Rehmannia glutinosa to scavenge 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)free radicals and hydroxyl radicals and their reducing ability were determined to evaluate the in vitro antioxidant activity of total polyphenols.The results showed that the optimal extraction method for total polyphenols from the root of Rehmannia glutinosa was a liquid-to-solid ratio of 10 mL/g,an ethanol concentration of 83%,an extraction temperature of 68℃,and an extraction time of 2.3 h.At this time,the total polyphenol extraction rate was 16.131%.The maximum scavenging rates of DPPH free radicals and hydroxyl radicals by the total polyphenols from the root of Rehmannia glutinosa were 85.94%and 60.81%,respectively,and the reducing ability was good.The study demonstrated that the antioxidant activity of the total polyphenols of the root of Rehmannia glutinosa is strong and can be used as a natural green antioxidant in animal production.
