特应性皮炎患者皮肤微生物组特征:基于16S rDNA测序分析

摘要 目的比较特应性皮炎(AD)患者与健康人群的皮肤微生物组成与多样性差异,为理解AD的微生物相关分子机制提供线索。方法收集81例AD患者(AD组)和63例健康皮肤(对照组)的皮肤样本,使用CTAB法提取DNA,进行16S rDNA测序,采用α多样性、β多样性和PCoA分析皮肤菌群差异。对皮肤菌群的组成进行门水平和属水平分析,将门和属水平前30丰度值的细菌进行热图分析。结果AD组与对照组在微生物组成上有差异。与对照组相比,AD组厚壁菌门和葡萄球菌相对丰度升高,放线菌门、变形菌门、红球菌属和乳杆菌属相对丰度降低。结论AD患者与健康人的皮肤菌群存在明显差异,需要进一步的研究来阐明微生物生态失调机制,为AD的靶向干预治疗铺平道路。 Objective To compare the differences in skin microbial composition and diversity between patients with atopic dermatitis and healthy individuals,to provide clues for understanding the microbe-related molecular mechanisms of AD.Methods Skin samples were collected from 81 patients with atopic dermatitis(AD group)and 63 healthy individuals(control group).DNA was extracted using the CTAB method,followed by 16S rDNA sequencing.Differences in skin microbiota were analyzed using alpha diversity,beta diversity,and principal coordinates analysis(PCoA).The composition of the skin microbiota was examined at the phylum and genus levels,and heatmaps were generated for the top 30 most abundant bacteria at both levels.Results There were differences in the microbiome between the control group and the AD group.Compared with the control group,the AD group showed increased relative abundance of Firmicutes and Staphylococcus,while Actinobacteria,Proteobacteria,Rhodococcus,and Lactobacillus showed decreased relative abundance.Conclusions There are significant differences in the skin microbiota between patients with AD and healthy individuals.Further research is needed to elucidate the mechanisms of microbial dysbiosis,paving the way for targeted therapeutic interventions for AD.
