摘要
目的优化龙胆DNA提取方法,建立一种针对龙胆的特异性聚合酶链式反应(polymerase chain reaction,PCR)鉴别方法,快速、准确地鉴别龙胆及其伪品。方法改良CTAB法提取龙胆及其伪品DNA,设计龙胆特异性引物,优化PCR反应体系,并对龙胆及伪品进行特异物种PCR鉴定和方法学考察。结果改良CTAB法后,提取的龙胆DNA浓度及纯度显著提高,特异性引物可区分龙胆及其伪品,灵敏度高、重复性好。结论该方法简便快速,可为龙胆鉴别提供参考。
Objective To optimize the DNA extraction method for Gentiana scabra Bunge,establish a specific poly-merase chain reaction(PCR)identification method for Gentiana scabra Bunge,and identify Gentiana scabra Bunge and its adulterants rapidly and accurately.Methods The modified CTAB method was used to extract the DNA of Gentiana scabra Bunge and its adulterants.Specific primers of Gentiana scabra Bunge were designed,and the PCR reaction system was optimized.The specific species of Gentiana scabra Bunge and its adulterants were identified by PCR and methodological validation was performed.Results The concentration and purity of Gentiana scabra Bunge DNA extracted by modified CTAB method were significantly improved,and the specific primers could distinguish Gentiana scabra Bunge and its adulterants with high sensitivity and good repeatability.Conclusion The method is simple and rapid,and can provide a reference for identification of Gentiana scabra Bunge.
