摘要
以29个酸枣品种(系)为试材,采用荧光毛细管电泳技术对其进行基因分型,研究了酸枣种质资源遗传多样性与种质亲缘关系,以期为辽宁酸枣核心种质构建提供参考依据。结果表明:12对SSR引物共检测到69个等位基因(Na),每对引物检测到2~8个Na。有效等位基因(Ne)在1.646~4.816,共有39.936个,占57.88%。Shannon信息指数(I)变幅介于0.552~1.668,平均值为1.258。观测杂合度(Ho)介于0.186~0.972,平均值为0.670。期望杂合度(He)介于0.372~0.789,平均值为0.663。不同引物揭示的多态信息含量(PIC)在0.372~0.835,平均为0.708。遗传相似系数在0~1,平均为0.28。聚类分析结果表明,在遗传距离为0.32时,可将29份酸枣种质分为四大类群。构建了酸枣指纹图谱及分子身份证,除2个品系外均具有唯一性。构建酸枣指纹图谱切实可行,可为酸枣品种身份识别、溯源管理及种质创新提供技术支撑和参考依据。
Taking 29 Z.jujuba varieties(lines)as the test materials,fluorescence capillary electrophoresis technology was used to genotype the genetic diversity and genetic relationships of Ziziphus jujuba germplasm resources was explored,in order to provide reference for the construction of core germplasm of Liaoning Ziziphus jujuba.The results showed that a total of 69 alleles(Na)were detected by 12 pairs of SSR primers,with 2-8 Na detected by each primer pair.The effective allele(Ne)ranged from 1.646 to 4.816,with a total of 39.936,accounting for 57.88%.The Shannon information index(I)ranged from 0.552 to 1.668,with an average of 1.258.The observed heterozygosity(Ho)ranged from 0.186 to 0.972,with an average of 0.670.The expected heterozygosity(He)ranged from 0.372 to 0.789,with an average of 0.663.The polymorphic information content(PIC)revealed by different primers ranged from 0.372 to 0.835,with an average of 0.708.The genetic similarity coefficient ranged from 0 to 1,with an average of 0.28.At the genetic distance of 0.36,the 29 Z.jujuba materials were divided into four categories by cluster analysis.Based on these results,we constructed a fingerprint and molecular ID card of Z.jujuba,which was unique to all but two strains.The construction of Z.jujuba fingerprint map was feasible and could provide technical support and reference basis for the identification,traceability management,and germplasm innovation of Z.jujuba varieties.
