mPEG-PLA-PHis-ss-OEI嵌段共聚物的合成 及体外生物安全性研究

Synthesis and in Vitro Biological Safety Study of mPEG-PLA-PHis-ss-OEI Block Copolymer

摘要 采用市售的共聚物原料,设计并合成了嵌段共聚物mPEG-PLA-PHis-ss-OEI(PPP-ss-O),采用核磁共振波谱法表征化学结构,凝胶渗透色谱法测定相对分子质量,比荧光探针法测定临界胶束溶度值,使用兔血细胞考察嵌段共聚物的溶血率、CT26肿瘤细胞验证其细胞毒性。结果表明PPP-ss-O表征图谱与预期结构一致,GPC呈单峰分布,分散系数(PDI)较小,相对分子质量为7640,临界胶束含量为2.4×10-6,溶血试验和细胞活力检测结果均显示,与聚乙烯亚胺(OEI)相比,PPP-ss-O具有良好的生物安全性。PPP-ss-O的成功合成和良好的体外生物安全性,为后续PPP-ss-O用于核酸类药物递送、构建纳米递送系统及相关应用打下了坚实的材料基础。 The block copolymer mPEG-PLA-PHis-ss-OEI(PPP-ss-O)was designed and synthesized.The chemical structure was characterized by nuclear magnetic resonance spectroscopy,the molecular weight was determined by gel permeation chromatography,the critical micelle concentration was measured by fluorescence probe method,and the hemolysis rate of the block copolymer and the cytotoxicity of CT26 tumor cells were verified by rabbit blood cells.The results showed that the characterization graph of PPP-ss-O was consistent with the expected structure.The GPC showed a single peak distribution,with a small dispersity index(PDI),a molecular weight of 7640,and a critical micelle concentration of 2.4×10-6.The hemolysis test and cell viability detection results indicated that compared with OEI,PPP-ss-O had good biocompatibility.The successful synthesis of PPP-ss-O and its good in vitro biocompatibility laid a solid material foundation for its subsequent application in nucleic acid drug delivery,construction of nano-delivery systems,and related applications.
