摘要
目的 建立同时测定菊花决明子茶中8种有效成分(绿原酸、咖啡酸、阿魏酸、毛蕊花糖苷、木犀草苷、芦丁、槲皮素及橙黄决明素)的高效液相色谱(high performance liquid chromatography,HPLC)分析方法。方法 采用Waters HSS T3 C_(18)色谱柱(250 mm×4.6 mm,5μm),以甲醇-0.1%磷酸溶液为流动相进行梯度洗脱(0~20 min:10%~30%甲醇;20~30 min:30%~60%甲醇;30~40 min:60%~10%甲醇;40~50 min:10%甲醇),流速1.0 mL/min,柱温30℃,检测波长320 nm。结果 8种成分在对应的浓度区间内呈现出良好的线性关系(r≥0.9968),平均加样回收率范围为87.02%~101.95%,相对标准偏差0.65%~1.39%。结论 该方法准确可靠,适用于菊花决明子茶的质量控制,分析结果显示不同批次产品中抗氧化成分含量存在显著差异。
Objective To establish an high performance liquid chromatography(HPLC)method for the simultaneous determination of 8 kinds of active components(chlorogenic acid,caffeic acid,ferulic acid,verbascoside,luteoloside,rutin,quercetin and aurantio-obtusin)in chrysanthemum and cassia seed tea.Methods Separation was performed on a Waters HSS T3 C18 column(250 mm×4.6 mm,5μm)using a gradient elution of methanol-0.1%phosphoric acid solution(0–20 min:10%–30%methanol;20–30 min:30%–60%methanol;30–40 min:60%–10%methanol;40–50 min:10%methanol)at a flow rate of 1.0 mL/min.The column temperature was maintained at 30℃with detection wavelength set at 320 nm.Results The 8 kinds of components showed good linear relationships within their respective concentration ranges(r≥0.9968).The average recoveries ranged from 87.02%to 101.95%with relative standard deviations of 0.65%–1.39%.Conclusion The developed method is accurate and reliable for quality control of chrysanthemum and cassia seed tea,revealing significant variations in antioxidant component content among different batches.
