猫传染性腹膜炎的RT-PCR诊断及病理组织学观察

摘要 本文对两例疑似感染猫传染性腹膜炎病毒的死亡患猫,通过观察临床症状、实验室检查、病原RT-PCR检测以及病理组织切片等方法进行确诊。首先,收集患猫腹水样品进行RT-PCR检测呈阳性,并构建系统发育树、进行同源性分析,显示两猫腹水样品均能扩增出417 bp大小的阳性目的条带,序列与猫冠状病毒有很高的同源性。系统发育树显示,与猫传染性腹膜炎病毒处于同一发育分支;再次通过病理组织切片镜检结果显示肝脏、脾脏、肾脏等被膜外均有大量纤维素渗出,并伴有炎性细胞、广泛性脂肪变性,心肌纤维发生颗粒变性等严重病理损伤。最终确诊2例患猫均感染了猫传染性腹膜炎病毒,且该病毒对猫多种主要器官造成了严重损伤。 This study diagnosed two suspected cases of death cats infected with feline infectious peritonitis virus through clinical symptoms,laboratory examination,pathogen RT-PCR detection and pathological tissue slices.Firstly,the ascitic fluid samples of the diseased cats were collected for RT-PCR detection and showed positive results.The phylogenetic tree was constructed and homology analysis was carried out.The results showed that the ascitic fluid samples of the two cats could amplify the positive target strip with the of 417 bp.The sequence had a high homology with the feline coronavirus,and the phylogenetic tree showed that it was in the same branch of development of the feline infectious peritonitis virus.Then,the pathological tissue slice microscopic examination results showed that there was a large amount of fibrin exudation outside the of the liver,spleen and kidney,accompanied by inflammatory cells,extensive fatty degeneration,granular degeneration of myocardial fibers and other serious pathological damage,the two cases of diseased cats were diagnosed with infection of feline infectious peritonitis virus,and the virus had caused serious damage to a variety of major organs of cats.
