基于甲状腺-肾上腺轴针刺强壮穴对HT大鼠ACTH、TSH、CORT、ALD水平的影响

Effect of acupuncture at Qiangzhuang points based on thyroid-adrenal axis on ACTH,TSH,CORT,ALD levels in HT rats

摘要 目的通过针刺强壮穴干预桥本氏甲状腺炎(HT)大鼠甲状腺功能水平,观察ACTH、CORT、ALD的含量,探讨甲状腺-肾上腺轴之间的相关性以及对肾上腺皮质功能的影响。方法雌性SD大鼠18只,随机选取6只为空白组,其余大鼠均制成HT模型,造模成功后随机分为模型组和针刺组各6只。采用ELISA法检测大鼠血清TSH、ACTH、FT3、FT4、CORT、TGAb、TPOAb、ALD含量,观察肾上腺组织病理学变化,采用Pearson法分析TSH水平与ACTH、CORT、ALD水平的相关性。结果与空白组比较,模型组大鼠血清TPOAb、TGAb、FT3、FT4含量均有不同程度升高(P<0.01),ACTH、TSH、CORT、ALD含量均有不同程度降低(P<0.01);与模型组比较,针刺组大鼠血清ACTH、TSH、CORT、ALD含量均有不同程度上升(P<0.01),TPOAb、TGAb、FT3、FT4含量均有不同程度下降(P<0.01)。Pearson法分析结果显示,HT大鼠血清TSH与ACTH、CORT、ALD水平呈正相关(P<0.05)。光镜下模型组大鼠肾上腺细胞大小不一,体积增大,核呈类圆形,胞浆空亮,呈透明样空泡状,形状不规则;针刺组大鼠胞浆内未见明显透明样空泡。结论针刺HT大鼠强壮穴显示血清TSH水平与ACTH、CORT、ALD水平存在正相关性,对甲状腺及肾上腺皮质功能的恢复有相关性。 Objective Acupuncture at Qiangzhuang points was used to interfere with thyroid function in HT rats,and the contents of ACTH,CORT,and ALD were observed to explore the correlation between thyroid-adrenal axis and its influence on adrenal cortex function.Methods Eighteen female SD rats were randomly selected and 6 rats were randomly selected as blank group.The rest rats were made into HT models.After successful modeling,they were randomly divided into model group and acupuncture group with 6 rats in each group.The serum TSH,ACTH,FT3,FT4,CORT,TGAb,TPOAb,and ALD contents of rats were measured by ELISA,and the histopathological changes of adrenal gland were observed.The correlation between TSH levels and ACTH,CORT,and ALD levels was analyzed by Pearson method.Results Compared with the blank group,the serum contents of TPOAb,TGAb,FT3,and FT4 in rats in the model group were increased to varying degrees,and the contents of ACTH,TSH,CORT,and ALD were decreased to varying degrees(P<0.01);Compared with the model group,the serum contents of ACTH,TSH,CORT,and ALD in rats in the acupuncture group were increased to varying degrees,and the contents of TPOAb,TGAb,FT3,and FT4 were decreased to varying degrees(P<0.01).Pearson's method analysis showed that serum TSH was positively correlated with the levels of ACTH,CORT,and ALD in HT rats(P<0.05).Under the light microscope,the adrenal cells in the model group were of different sizes and increased in volume,the nuclei were round,the cytoplasm was empty and bright,and they were transparent vacuol-like,with irregular shapes.There were no obvious transparent vacuol-like vacuol-like in the cytoplasm of the rats in the acupuncture group.Conclusion Acupuncture at Qiangzhuang points in HT rats showed a positive correlation between serum TSH levels and ACTH,CORT,and ALD levels,and was correlated with the recovery of thyroid and adrenal cortex functions.
