基于网络药理学从自噬视角探讨参苓白术散治疗膜性肾病的作用机制

摘要 目的基于网络药理学探讨参苓白术散治疗膜性肾病的作用机制。方法通过检索TCMSP数据库(检索时间截止到2024年6月30日),收集参苓白术散的活性成分及相关靶点,在GenaCards、DrugBank、OMIM数据库中收集膜性肾病疾病靶点,通过构建“药物-成分-靶点”的复杂网络,获取参苓白术散的核心成分;然后使用STRING平台进行PPI分析;最后运用DAVID数据库进行GO和KEGG富集分析,并通过微生信平台进行可视化处理。结果共获取中药复方参苓白术散活性成分190个,去重后获得相关靶点286个,膜性肾病疾病靶点1499个,药物与疾病的交集基因157个。通过构建靶点PPI,筛选出10个核心靶点。GO富集获得934条生物过程(BP),102条细胞组分(CC),146条分子功能(MF);KEGG主要富集在185等条信号通路。结论参苓白术散通过多成分、多靶点、多通路治疗膜性肾病。 Objective To explore the mechanism of Shenling Baizhu Powder in treating membranous nephropathy based on network pharmacology.Methods The active components and related targets of Shenling Baizhu Powder were collected by searching TCMSP database(Search time is up to June 30,2024),and the targets of membranous nephropathy disease were collected from GenaCards,DrugBank and OMIM databases.The core components of Shenling Baizhu Powder were obtained by constructing a complex network of"drugingredient-target".Then the STRING platform was used for PPI analysis.Finally,GO and KEGG enrichment analysis was carried out using DAVID database,and visualization processing was carried out by Weisheng platform.Results A total of 190 active components of Shenling Baizhu Powder were obtained,286 related targets,1499 MN disease targets and 157 crossover genes between drugs and diseases were obtained after weight removal.By constructing target PPI,10 core targets were selected.GO enrichment obtained 934 biological processes(BP),102 cell components(CC)and 146 molecular functions(MF).KEGG is mainly concentrated in 185 signal pathways.Conclusions Shenling Baizhu Powder is a multi-component,multi-target and multi-pathway treatment for membranous nephropathy.
