摘要
目的:建立同时测定诃子汁中没食子酸、表没食子儿茶素没食子酸酯、诃黎勒酸、诃子酸和鞣花酸5种多酚类化合物的HPLC法,并测定不同pH条件下诃子汁中五种多酚类化合物的变化。方法:采用Agilent Zorbax C_(18)柱(250 mm×4.6 mm,5μm),流动相为甲醇(A)-0.1%磷酸水(B),梯度洗脱,流速1.0 mL/min,柱温25℃,紫外检测器检测波长为270 nm,进样量5μL。结果:诃子汁中5种多酚类化合物分离度良好,在一定范围内呈现出良好的线性关系,精密度、重复性良好,在室温条件下24h内稳定,平均加样回收率在100.07%~102.37%之间(RSD为1.25%~3.24%)。不同pH条件对5种多酚类化合物含量都有影响,其中pH条件的不同对诃子酸含量的影响最小,对没食子酸含量的影响最大。结论:该试验方法简单准确,重复性好,可用于同时测定没食子酸、EGCG、诃黎勒酸、诃子酸和鞣花酸成分,并测定不同pH下五种化合物含量的变化,为诃子汁的质量控制和加工贮藏工艺提供了一定的理论依据。
Objective An HPLC method was established for the simultaneous determination of five polyphenolic compounds,gallic acid,epigallocatechin gallate(EGCG),chebulagic acid,chebulinic acid,and ellagic acid,in the Chebulae juice,and for the deter-mination of the changes of the five polyphenolic compounds in the Chebulaejuice under different pH conditions.Methods The separa-tion was performed on an Agilent Zorbax C_(18) column(250 mm×4.6 mm,5μm)with the mobile phase of methanol(A)-0.1%phosphoric acid in water(B)in a gradient elution at a flow rate of 1.0 mL/min,the column temperature was 25℃,and the detec-tion wavelength of the UV detector was 270 nm with the injection volume of 5μL.Results The five polyphenolic compounds in Chebu-lae juice were well separated and showed good linear relationship within a certain range,with good precision and reproducibility,and were stable within 24 h at room temperature,and the average spiked recoveries ranged from 100.07%to 102.37%(RSDs of 1.25%-3.24%).Different pH conditions had an effect on the contents of all five polyphenolic compounds,in which the difference of pH conditions had the least effect on the content of chebulinic acid and the most effect on the content of gallic acid.Conclusion The test method is simple,accurate and reproducible,and can be used for the simultaneous determination of gallic acid,EGCG,chebulagic acid,chebulinic acid and ellagic acid components,and the determination of the changes in the content of the five compounds at differ-ent pH,which provides a certain theoretical basis for the quality control of Chebulae juice and the processing and storage process.
