白绵毛兰组培快繁体系研究

摘要 以白绵毛兰蒴果为外植体,采用单因素试验设计,研究种子萌发试验、增殖试验、生根壮苗试验。结果表明,白绵毛兰蒴果中种子萌发率最高月份为11月;种子萌发最优培养基为1/2 MS+25 g/L糖+8 g/L卡拉胶+25 g/L马铃薯粉+0.1 mg/L NAA和1/2 MS+25 g/L糖+8 g/L卡拉胶+25 g/L马铃薯粉+0.1 mg/L NAA+0.1 g/L AC,种子最高萌发率达70%;增殖最佳激素为6-BA,NAA和2,4-D不宜用于增殖;在不切除组培小苗苗根光照培养条件下,最适增殖培养基为1/2 MS+25 g/L糖+8 g/L卡拉胶+2 mg/L 6-BA,继代周期为90 d,小苗不定芽增殖系数达3.28;在不切除组培小苗苗根弱光培养条件下,最适增殖培养基为1/2 MS+25 g/L糖+8 g/L卡拉胶+3 mg/L 6-BA,继代周期为90 d,小苗不定芽增殖系数达3.60;生根壮苗培养基添加30 g/L马铃薯粉对芽的生长有促进作用,平均芽数达2.43;生根壮苗培养基添加20 g/L香蕉粉对根的生长有显著影响,平均根长达2.42 cm;最适生根壮苗组合培养基为1/2 MS+25 g/L糖+8 g/L卡拉胶+30 g/L马铃薯粉+20 g/L香蕉粉。该研究旨在建立白绵毛兰组培快繁技术体系,保存白绵毛兰种质资源,为今后开发利用提供一定的基础材料及技术参考价值。 The capsule of Dendrolirium lasiopetalum was used as explants,and the seed germination test,proliferation test and rooting seedling test were carried out by single factor experimental design.The results showed that the highest seed germination rate was in November.The optimal medium for seed germination was 1/2 MS+25 g/L sugar+8 g/L carrageenan+25 g/L potato powder+0.1 mg/L NAA and 1/2 MS+25 g/L sugar+8 g/L carrageenan+25 g/L potato powder+0.1 mg/L NAA+0.1 g/L AC.The maximum germination rate of seeds was 70%at 0.1 g/L AC.The best proliferative hormones were 6-BA,NAA and 2,4-D were not suitable for proliferation.The optimum culture medium was 1/2 MS+25 g/L sugar+8 g/L carrageenan+2 mg/L 6-BA.The subculture period was 90 days,the shoot bud multiplication coefficient of seedlings was 3.28;when the seedlings were cultured under weak light conditions without cutting the root,the optimal multiplication medium was 1/2 MS+sugar 25 g/L+agar 8 g/L+6-BA 3 mg/L,with a regeneration cycle of 90 days,and the shoot bud multiplication coefficient was 3.60;the addition of 30 g/L potato flour to the rooting and fattening medium had a significant effect on the shoot buds,with an average shoot of 2.43;the addition of 20 g/L banana flour to the rooting and fattening medium had a significant effect on root growth,with an average root coefficient of 2.42 cm;the optimal rooting and fattening medium combination was 1/2 MS+sugar 25 g/L+agar 8 g/L+30 g/L potato flour+20 g/L banana flour.It is hoped that a fast multiplication technology system for Dendrolirium lasiopetalum can be established,its genetic resources can be preserved,and a certain basis of material and technical reference value can be provided for future development and utilization.
